摘  要:目的探究慢性十二指肠炎症对帕金森病致病蛋白pSer129-α-syn(磷酸化的α突触核蛋白)积累的影响,为帕金森病的机制研究提供新的思路。方法利用给予野生型雄性Wistar大鼠口饲半胱氨酸盐酸盐的方法建立慢性十二指肠溃疡模型,利用十二指肠石蜡切片HE染色法评估十二指肠绒毛状态及炎性细胞浸润情况,利用十二指肠石蜡切片免疫组化法评价十二指肠局部致病蛋白pSer129-α-syn的聚集情况,采用脑部矢状切石蜡切片免疫组化法探讨脑部海马区致病蛋白pSer129-α-syn的聚集情况。结果长期慢性十二指肠溃疡能导致致病蛋白pSer129-α-syn在十二指肠局部少量聚集,也可以引起脑部致病蛋白pSer129-α-syn的积累。结论长期慢性十二指肠溃疡可以增加帕金森病的致病蛋白pSer129-α-syn在肠道及大脑中的积累。Objective To explore the influence of chronic duodenal inflammation on the accumulation of pathogenic protein pSer129-α-syn in Parkinson′s disease, and provide a new idea for the exploration of the source of pathogenic protein in Parkinson′s disease. Methods The rat models of chronic duodenal ulcer were established and divided into a control group and ulcer group(5 rats in each group). HE staining of paraffin section of the duodenum was used to evaluate the intestinal villus status and inflammatory cell accumulation. Immunohistochemistry of the paraffin section of duodenum was used to evaluate the aggregation of local pathogenic protein pSer129-α-syn. Immunohistochemistry of paraffin sections in the sagittal section of brain was used to observe the accumulation of pathogenic protein pSer129-α-syn in the hippocampus. Results Chronic duodenal ulcer could lead to accumulation of a small amount of pathogenic protein pSer129-α-syn in the duodenum, and also accumulation of pathogenic protein pSer129-α-syn in the brain. Conclusion Chronic duodenal ulcers can increase the accumulation of the pathogenic protein pSer129-α-syn in the duodenum and brain.
