摘  要:目的 探讨纳洛酮对内吗啡肽-1(EM-1)减轻大鼠心肌缺血再灌注损伤作用的影响。方法 将30只SD雄性大鼠随机分为假手术组(S组)、心肌缺血再灌注组(IR组)、内吗啡肽-1后处理组(EM50组)、内吗啡肽-1+纳洛酮后处理组(EM50+Nal组)、纳洛酮后处理组(Nal组);S组冠状动脉左前降支(LAD)下穿丝线不结扎维持150 min, EM50组在结扎LAD 25 min后经尾静脉输注50μg·kg^(-1)EM-1,EM50+Nal组结扎LAD 10 min后静脉推注3 mg·kg^(-1)纳洛酮,在灌注前5 min将经静脉输注50μg·kg(-1)EM-1,Nal组结扎LAD 10 min后,静脉注射3 mg·kg^(-1)纳洛酮。用酶联免疫吸附试验试剂盒法检测血浆中乳酸脱氢酶(LDH)、心肌肌钙蛋白Ⅰ(cTnⅠ)、肌酸激酶(CK-MB)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)以及丙二醛(MDA)的含量。结果 S组、IR组、EM50组、EM50+Nal组和Nal组的血浆中CK-MB表达量分别为(38.47±3.88),(76.27±4.25),(41.48±2.42),(70.33±2.50)和(67.58±3.89) U·L^(-1);这5组血浆中TNF-α表达量分别为(123.58±12.06),(200.70±7.42),(135.50±5.04),(158.25±7.72)和(184.16±9.96) pg·mL^(-1);这5组血浆中SOD表达量分别为(143.86±7.41),(84.10±12.42),(132.77±8.25),(88.19±7.92)和(94.70±18.31) U·mL^(-1)。IR组的上述指标与EM50组相比,差异均有统计学意义(均P<0.05);EM50组的上述指标与EM50+Nal组相比,差异均有统计学意义(均P<0.05)。结论 纳洛酮抑制了内吗啡肽-1后处理对心肌缺血再灌注损伤的保护作用。ObjectiveTo investigate the effect of naloxone on endomorphin-1 (EM-1) in reducing myocardial ischemiareperfusion injury in rats.Methods Thirty SD male rats were randomly divided into sham operation group (S group),myocardial ischemia reperfusion group (IR group),endomorphin-1 post-treatment group(EM50 group),endomorphin-1+naloxone post-treatment group(EM50+Nal group),naloxone post-treatment group (Nal group).The left anterior descending coronary artery (LAD) of rats in S group was not ligated for 150 min;EM50 group received intravenous injection of 50μg·kg(-1)EM-1 after 25 min of LAD;EM50+Nal group received intravenous injection of 3 mg·kg^(-1) naloxone after 10 min of ligation of LAD,and intravenous injection of 50μg·kg(-1)EM-1 in 5 min before reperfusion;Nal group received intravenous injection of 3 mg·kg^(-1) naloxone after 10 min of ligation of LAD.Separation of plasma using the enzyme-linked immunosorbent assay kit to detect lactate dehydrogenase (LDH),cardiac troponin I (cTnI),creatine kinase-MB(CK-MB),interleukin-6 (IL-6),tumor necrosis factor-α (TNF-α),super-oxide dismutase (SOD) and malondialdehyde (MDA).Results The expression levels of CK-MB in plasma of S group,IR group,EM50 group,EM50+Nal group and Nal group were (38.47±3.88),(76.27±4.25),(41.48±2.42),(70.33±2.50) and(67.58±3.89) U·L^(-1);the expression of TNF-α in the plasma of these five groups were (123.58±12.06),(200.70±7.42),(135.50±5.04),(158.25±7.72) and (184.16±9.96) pg·mL^(-1);the expression of SOD in plasma of these five groups were (143.86±7.41),(84.10±12.42),(132.77±8.25),(88.19±7.92) and(94.70±18.31) U·mL^(-1).In the above indicators,the differences between the IR group and the EM50 group were statistically significant (all P<0.05);the differences between the EM50 group and the EM50+Nal group were statistically significant (all P<0.05).Conclusion Naloxone inhibited the protective effect of endomorphin-1 posttreatment on myocardial ischemia-reperfusion injury.
