摘  要:采用微填充床反应器首次实现了利普斯他汀连续流动加氢合成奥利司他,优化的反应条件为:氢气压力为1 MPa,温度为50℃,溶剂是庚烷,原料浓度是50 mg/mL,氢气/利普斯他汀摩尔比为3∶1,质量空速为0.3 h^(-1),催化剂采用1%Pd/Al_(2)O_(3)。相较于传统高压加氢釜工艺,连续流动加氢工艺节约钯催化剂80%,产率提升5%,同时具有本质安全的特点,更适合现代工业生产。Orlistat is synthesized through continuous flow hydrogenation of Lipstatin in a micro packed-bed reactor for the first time.The optimized conditions are as follows: hydrogen pressure is 1 MPa, reaction temperature is 50℃,heptane is used as solvent, the concentration of Lipstatin in heptane is 50 mg·mL,the molar ratio of Hto Lipstatin is 3∶1,mass space velocity is 0.3 h^(-1),and 1% Pd on Al_(2)O_(3)is used as catalyst.Compared with traditional high-pressure autoclave hydrogenation process, this continuous flow hydrogenation process can save 80% of catalyst, and increase the yield by 5%.This inherently safe continuous flow hydrogenation process is more suitable for modern industrial production.
