摘 要:以佛波酯(PMA)于体外培养中刺激原代法氏囊细胞(PBCs)诱导产生的IgM^(+)TIM4^(+)B细胞为模型,分析传染性囊病病毒(IBDV)感染对IgM^(+)TIM4^(+)B细胞分化形成及IBDV在IgM^(+)TIM4^(+)B和IgM^(+)TIM4^(-)B细胞中的复制动态。结果显示,PMA刺激后第24~48小时PBCs中IgM^(+)TIM4^(+)B细胞比例相对稳定;而以IBDV感染经PMA刺激24 h的PBCs后,IgM^(+)TIM4^(+)B细胞比例显著减少且与IBDV感染剂量成负相关,IgM^(+)TIM4^(+)B细胞的IBDV-VP1 RNA水平也显著低于IgM^(+)TIM4^(-)B细胞。结果提示,IBDV在IgM+TIM4+B细胞和IgM^(+)TIM4^(-)B细胞中的复制过程存在差异。Here we developed a cellular model in vitro of IgM^(+)TIM4^(+)B cell induced from the primary bursa cells(PBCs)by the phorbol myristate acetate(PMA)treatment for probing the impacts of infectious bursal disease virus(IBDV)on the formation of IgM^(+)TIM4^(+)B cells,and the replications of IBDV in this novel B cell subset.The results revealed that an unfluctuating proportion of IgM^(+)TIM4^(+)B cells could be induced in PBCs during the 24—48 hours after PMA stimulation.However,the PBCs treated with PMA for 24 h were infected with IBDV,the ratio of IgM^(+)TIM4^(+)B cells was significantly reduced and negatively correlated with the infection dose of IBDV,and simultaneously the RNA level of IBDV-VP1 in IgM^(+)TIM4^(+)B cells was significantly decreased compared to that of IgM^(+)TIM4^(-)B cells.These results suggested the differences in IBDV replication in IgM^(+)TIM4^(+)B cells and IgM^(+)TIM4^(-)B cells.
