摘 要:目的 基于二代测序平台对生物检材的微单倍型遗传标记进行检验,并探讨微单倍型遗传标记在法医物证学中的应用价值。方法 使用MHseq Typer47微单倍型试剂制备文库,MiSeq FGx平台进行测序,MHTyper微单倍型数据分析软件进行数据分析。结果 47个微单倍型位点经二代测序全部检出,灵敏度达60 pg,1:19的混合样品可以准确拆分,法医物证检材的检验结果良好。结论 微单倍型遗传标记在法医物证检验中具有较高的应用价值。Objective To obtain microhaplotype loci in forensic biological evidence with next-generation sequencing and explore its prospects in forensic genetics application. Methods The MHseq Typer47 microhaplotype kit was used for library preparation, and massively parallel sequencing was performed on the MiSeq FGx system. The MHTyper Microhaplotype Data Analysis Software was used for data analysis. Results 47 microhaplotype loci could be typed accurately with a sensitivity of 60 pg, the mixed profiles could be separated at the ratio of 1:19;the results were satisfactory.Conclusion The microhaplotypic genetic markers are valuable for the practice of forensic genetics.
