摘  要:目的 比较纯水和牙膏作为两步擦拭法的润湿介质提取接触性DNA的分型检出效果。方法 分别用纯水和牙膏作为润湿介质,以干湿两步法擦拭提取同一个案件同一类型的不同客体上接触性DNA,磁珠法试剂盒提取并纯化接触性DNA,用Globalfiler试剂盒进行STR分型检验,比较使用两种润湿介质的检材STR分型检出率。结果牙膏作为润湿介质的检材检出率高于纯水,差异具有统计学意义(P <0.05)。结论 使用牙膏作为润湿介质两步擦拭法提取现场接触性DNA检材的效果优于纯水。Objective To compare the effectiveness of toothpaste and pure water as swabbing media for the detection of STR genotypes when analyzing touch DNA. Methods Touch DNA samples were swabbed off from different touched objects of the same type in the same criminal case, using double-swab technique with two swabbing media(pure water and toothpaste) respectively. The samples were extracted and purified by magnetic bead method, amplified and genotyped by Globalfiler Kit. The STR genotypes detection rates of the testing material using the two swabbing media were compared.Results The detection rates of STR genotypes using toothpaste as swabbing medium was higher than that of pure water, the differences had statistical significance(P < 0.05). Conclusion The use of toothpaste as swabbing medium to collect touch DNA is more effective.
