摘 要:通过使用脂多糖(LPS)制造小鼠肾炎模型,观察哈蟆油水提物对其肾脏的保护作用.选取42只小鼠,分为6组,即哈蟆油高剂量组、哈蟆油中剂量组、哈蟆油低剂量组、空白对照组、阳性对照组、模型组,每组7只.与空白对照组相比,模型组LPS能显著提高炎症及免疫因子IL-6、IL-1β、IFN-γ 和IgG含量(P<0.05),与模型组相比,灌胃哈蟆油水提物能显著降低炎症因子IL-6、IL-1β、IFN-γ含量,灌胃哈蟆油水提物低剂量组能显著降低免疫因子IgA和IgG含量(P<0.05);与空白对照组相比,模型组LPS能显著提高小鼠肾脏炎症相关基因NLRP3、NLRP1a、IL-1β的相对表达量(P<0.05),与模型组相比,灌胃哈蟆油水提物能显著降低肾脏炎症相关基因NLRP3、NLRP1 a、IL-1β相对表达量(P<0.05);与空白对照组相比,模型组LPS能显著上调小鼠体内NO、SOD和CAT的水平(P<0.05),与模型组相比,阳性组LPS能显著下调小鼠体内NO、SOD和CAT的水平(P<0.05),灌胃哈蟆油水提物高剂量组和低剂量组的小鼠能显著降低肾脏中抗氧化指标NO、CAT和SOD的水平(P<0.05).结果显示,哈蟆油对LPS诱导的肾脏炎症有着一定的保护作用,且在25 mg/kg作用下具有最佳的保护效果.The nephritis model of mice was made by LPS,and the protective effect of Oviductus ranae water extract on kidney was observed.Forty two female mice were selected and divided into 6 groups with 7 mice in each group,namely Oviductus ranae high-dose,medium-dose,low-dose groups,negative control group,positive control group and model group,respectively.Compared with the negative control group,LPS could significantly increase the contents of the inflammatory and immune factors of IL-6 and IL-1β,IFN-and IgG(P<0.05)of mice in the model group.Compared with the model group,intragastric administration of Oviductus ranae water extract could significantly reduce the contents of inflammatory factors of IL-6 and IL-1β,IFN-.The contents of IgA and IgG were significantly decreased in the low-dose group(P<0.05).Compared with the negative control group,LPS could significantly increase the renal inflammation gene relative expression of NLRP3,NRPla and IL-1βof mice in the model group.But compared with the model group,intragastric administration of Oviductus ranae water extract could significantly reduce these gene relative expression(P<0.05).Compared with the negative control group,LPS could significantly up regulate the levels of NO,SOD and CAT of mice in the model group(P<0.05).Compared with the model group,LPS could significantly down regulate the levels of NO,SOD and CAT of mice in the positive group(P<0.05),and Oviductus ranae water extract high-dose and low-dose groups could significantly reduce the levels of antioxidant indexes of NO,CAT and SOD in kidney of mice(P<0.05).The results showed that Oviductus ranae had a certain protective effect against LPS induced renal inflammation,and had the best protective effect at 25 mg/kg.
