摘  要:目的:探讨栀子豉汤治疗反流性食管炎(reflux esophagitis,RE)潜在的作用机制。方法:通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索栀子、淡豆豉的活性成分以及作用靶点,再通过靶标数据库(therapeutic target database,TTD)、人类基因数据库(GeneCardsⓇ:the human gene database)、在线人类孟德尔遗传数据库(online mendelian inheritance in man,OMIM)、PharmGkb、DrugBank等5个数据库检索反流性食管炎的靶标,取并集;利用Venn在线程序将药物成分与疾病靶点去重并取交集,在Cytoscape3.8.0软件中进行可视化分析,构建药物疾病靶点网络图;再利用String在线网站,及Cytoscape 3.8.0筛选出核心蛋白互作网络,构建蛋白质相互作用(protein-protein interaction,PPI)网络图;最后利用R 4.1.2软件以P<0.05作为条件,对核心蛋白互作网络进行基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(kyoto encyclopedia of genes and ge-nomes,KEGG)通路富集分析。结果:共获得药物化学成分18个,药物作用靶点405个,疾病靶点2076个,药物作用靶点与疾病靶点交集108个,GO生物富集分析共获得涉及生物学过程BP 1962个,细胞组分CC共48个,分子功能MF共154个,KEGG通路富集得出155条信号通路。结论:栀子豉汤中的各药物可能主要是通过调控机体血糖水平,以及抑制p38、NF-κB信号通路的激活,抑制炎性因子的分泌,从而达到治疗反流性食管炎的作用。Objective:To explore the potential mechanism of Gardenia and Semen Sojae Praeparatum Decoction in the treatment of reflux esophagitis(RE)based on network pharmacology.Methods:The active components and action targets of Zhizi(Fructus Gardeniae)and Dandouchi(Semen Sojae Praeparatum)were retrieved through TCMSP website,and then the targets of RE were retrieved through GeneCards,OMIM,PharmGkb,TTD and DrugBank,and unions were obtained.By using Venn online program,the weight of drug components and disease targets were removed and the intersection was taken,and the visual analysis was carried out in Cytoscape3.8.0 software to construct the network diagram of drug disease targets.Then,by using String online website and Cy-toscape 3.8.0,the core protein interaction network was screened out and the PPI network diagram was constructed.Finally,R4.1.2 software was used to analyze the GO biological enrichment and KEGG pathway enrichment of the core protein interaction network under the condition of P<0.05.Results:A total of 18 chemical components of drugs,405 drug action targets,2,076 disease tar-gets,108 intersection between drug action targets and disease targets were obtained.A total of 1,962 BP involving biological processes,48 cell component CC,and 154 molecular function MF were obtained by GO biological enrichment analysis.And 155 signal pathways were obtained by KEGG pathway enrichment.Conclusion:The drugs in Gardenia and Semen Sojae Praeparatum Decoction may inhibit the secretion of inflammatory factors mainly by regulating the blood glucose level of the body and inhibiting p38 and NF-κB activation of signal pathway so as to achieve the effect of treating RE.
