摘  要:为了研究小麦腺嘌呤转移酶基因SLC25A4在小麦生长发育中的生物学功能,本研究借助RT-PCR从‘贵紫麦1号’中获取SLC25A4-7A基因的cDNA序列全长,并对其编码的蛋白进行生物信息学分析。结果表明,SLC25A4-7A基因的开放阅读框为1137 bp,编码378个氨基酸,其编码的蛋白是一个亲水的稳定蛋白。SLC25A4-7A定位于线粒体膜,该蛋白的保守结构域与在ADP/ATP转运中起重要作用的PTZ00169序列相似。蛋白三级预测结果进一步表明该蛋白具有线粒体ADP/ATP转运载体的功能。多重序列比对和系统进化树分析的结果表明,SLC25A4-7A与拟南芥AAC3在生物学功能上更为接近。本研究为进一步探究该基因的生物学功能提供了理论依据。In this study,the cDNA sequence of adenine transferase gene SLC25A4-7A was obtained from’Guizi1’wheat by RT-PCR and the encoded protein was analyzed by bioinformatics in order to study the biological function of SLC25A4-7A in wheat growth and development.The results demonstrated that open reading frame of SLC25A4-7A gene,which encoding 378 amino acids,was 1137 bp.SLC25A4-7A protein was predicted to locate in mitochondrial membrane,was a hydrophilic stable protein.The conserved domain of SLC25A4-7A protein was specific matched to PTZ00169,which plays important role in ADP/ATP transport.Furthermore,tertiary structure result demonstrated that SLC25A4-7A protein possess function of mitochondrial ADP/ATP transporter.Meanwhile,the results of multiple sequence alignment and genetic phylogenetic tree analysis dedicated that biological function of SLC25A4-7A was closely related to Arabidopsis ACC3.This study provides a theoretical basis for further exploring the biological function of SLC25A4-7A gene.
