摘 要:油棕核壳厚度性状是影响油棕单株棕榈油产量的重要因子。SHELL基因是控制油棕核壳厚度性状的关键基因,其显隐性与核壳厚度性状类型高度相关。本研究根据已发布的油棕SHELL基因序列中的SNP位点,设计了一对特异性CAPS引物和限制性酶切位点,利用该标记对18份已知三种核壳厚度类型油棕叶片的基因组DNA进行PCR扩增和酶切检测,随后在70份油棕材料中进行验证。酶切结果表明,厚壳型油棕酶切产物包含2条条带(280和250 bp);无壳型油棕酶切产物包含1条条带(550 bp),与酶切前无差异;薄壳型油棕酶切产物包含3条条带(550,280和250 bp)。油棕核壳厚度性状类型分子鉴定结果与田间调查结果相一致。本研究利用基于油棕核壳厚度控制基因SHELL的CAPS分子标记方法,能准确鉴别厚壳型(Dura)、薄壳型(Tenera)和无壳型(Pisifera)三种核壳厚度类型的油棕材料,为油棕育种材料的早期鉴定与选择、提高杂交育种效率提供了有效途径。The shell thicknesses of oil palm is an important factor affecting the yield of individual oil palm.The SHELL gene is a key gene which control the shell thickness of oil palm,and its dominant and recessiveness are highly correlated with the type of shell thickness.In this study,a pair of CAPS primers and restriction site were designed based on the SNP in the published oil palm SHELL gene sequence.The primers were used to carry out PCR amplification and enzyme digestion on the genomic DNA of 18 oil palm leaves with three known shell thickeness.Subsequently,70 oil palm materials were applicated.The results showed that the dura enzyme digestion product consists of 2 bands(280 and 250 bp).The Pisifera enzyme digestion product consists of 1 bands(550 bp),the same as that before the enzyme digestion.The tenera enzyme digestion product consists of 3 bands(550,280 and 250 bp).The results of molecular identification is consistent with the field investigation of oil palm shell thickness types.In this study,we can accurately identify three types of oil palm materials with dura,tenera and pisifera,through the CAPS molecular marker based on SHELL gene.It provides an effective way for early identification and selection of oil palm breeding materials,improving the efficiency of hybrid breeding.
