摘 要:为构建高效的蒙古莸快繁体系来解决蒙古莸濒危的问题。本研究以蒙古莸新萌发幼嫩茎段为试验材料,采用3因素3水平正交试验设计和方差分析,利用组织培养技术构建蒙古莸组培快繁体系。本研究系统地分析了外植体的消毒方法及不同植物激素质量浓度对其不定芽诱导、增殖、生根过程中的作用及炼苗。研究结果表明:75%乙醇处理30 s配合以2%次氯酸钠处理3 min是蒙古莸外植体的最佳消毒方法,污染率为0;不定芽诱导与增殖中以MS+6-BA 1.0 mg/L+NAA 0.3 mg/L+GA_(3)0.02 mg/L条件下的芽诱导与芽增殖效果最好,诱导率达到最大为99%,增殖系数高达到12.00;生根诱导中以MS+IBA 0.5 mg/L+GA_(3)0.15 mg/L条件下的生根效果最好,生根率最高为89.5%;将生根苗移栽在草炭土:蛭石:珍珠岩=6:3:1的混合基质中,生活率达77%。本研究构建了一套完整的蒙古莸的组培快繁体系,本试验培育的组培苗的各项指标已达到快速繁育苗木的要求,其技术措施可直接应用于规模化生产,也为后期蒙古莸进一步的遗传转化研究提供理论基础。In o rder to construct an efficient reproducing system to solve the endangered problem of Caryopteris mongholica Bunge.Taking the young stem segments of C.mongholica Bunge as experimental materials,the tissue culture and rapid propagation system of C.mongholica Bunge was constructed by three factors and three levels orthogonal design and variance analysis.This study systematically analyzed the disinfection methods of explants and the effects of phytohormone on adventitious bud multiplication,rooting and seedling refining.The results showed that:The best sterilized method for the explants was to treat them with 75%ethanol for 30 s and 2%sodium hypochlorite for 3 min;in adventitious bud induction,the highest bud induction rate was 99%and the bud multiplication coefficient in adventitious bud multiplication reached 12.00 under MS+6-BA 1.0 mg/L+NAA 0.3 mg/L+GA_(3)0.02 mg/L;when the rooting seedlings of C.mongholica Bunge were transplanted into a mixed growth medium,in which the ratio of peat:perlite:vermiculite is 6:3:1,the survival rate was 77%.In this study,a complete rapid tissue culture system of C.mongholica Bunge was constructed.The indices of tissue culture seedlings cultivated in this experiment have met the requirements of rapid propagation of seedlings.Its technical measures can be directly applied to large-scale production,and provide a theoretical basis for further genetic transformation research of C.mongholica Bunge in the later stage.
