摘 要:为了探讨丹参素结合有氧运动对氧化途径及其机制影响,本研究选取60只雄性大鼠,设置6个研究组,通过不同剂量的处理,观察各组大鼠基本情况;测定丹参素结合有氧运动对各组大鼠骨密度、骨结构、骨质量、成骨分化能力等影响;分析大鼠肝组织中SOD、MDA含量,以及大鼠血清中GOT、GPT等指标;RT-qPCR检测Wnt途径相关基因表达。研究结果表明,与模型对照组相比,有氧运动组中血清SOD含量有所增加,经丹参素处理后,大鼠血清中SOD含量进一步增加,且呈剂量效应;与模型对照组相比,有氧运动组中血清MDA含量有所下降,经丹参素处理后,大鼠血清中MDA含量进一步下降,且呈剂量效应;与模型对照组相比,有氧运动组中大鼠血清中GOT、GPT等明显增加,经丹参素处理后,大鼠血清中GOT、GPT等含量进一步显著增加,且呈剂量效应;RT-qPCR检测各组大鼠肝脏中Wnt途径相关基因表达,结果发现丹参素结合有氧运动能够显著影响Wnt途径中Axin2和β-catenin基因的表达。有氧运动能够结合丹参素通过Wnt显著改善骨质疏松大鼠的状况。In order to explore the effect of Danshensu combined with aerobic exercise on the oxidation pathway and its mechanism,60 male rats were selected in this study,and six study groups were set up.The basic situation of rats in each group was observed through different doses of treatment;the effects of Danshensu combined with aerobic exercise on bone mineral density,bone structure,bone quality and osteogenic differentiation ability of rats in each group were measured;the contents of SOD and MDA in rat liver tissue and got and GPT in rat serum were analyzed;RT-qPCR was used to detect the expression of Wnt pathway related genes.Compared with the model control group,the level of SOD in the aerobic exercise group increased.After treatment with Danshensu,the level of SOD in the rat serum increased further.Compared with the model control group,the serum MDA content in the aerobic exercise group decreased,and after treatment with danshensu,the serum MDA content in the rats further decreased and showed a dose effect;compared with the model control group in the aerobic exercise group,GOT and GPT in the rat serum increased significantly.After treatment with Danshensu,the GOT and GPT contents in the rat serum increased significantly and showed a dose effect;RT-qPCR detected the expression of Wnt pathwayrelated genes in the liver of each group of rats.The results showed that aerobic exercise combined with Danshensu could significantly affect Axin2 andβ-catenin in the Wnt pathway.Aerobic exercise combined with Danshensu can significantly improve the condition of osteoporosis rats through Wnt.
