摘  要:目的探讨细胞因子信号转导抑制因子3(SOCS3)基因多态性与儿童支气管哮喘易感性的相关性。方法选取2017年1月—2020年1月安康市妇幼保健院门诊接收的106例支气管哮喘儿童为研究组,另选取同期来院体检的98例健康儿童为对照组。提取两组儿童外周血总DNA,检测SOCS3基因rs4969169位点基因型,比较两组SOCS3基因型和等位基因频率,分析研究组中SOCS3基因rs4969169位点与一般情况资料的关系,分析SOCS3基因rs4969169位点基因型与儿童支气管哮喘易感性的关系。结果研究组SOCS3基因rs4969169位点TT基因型频率高于对照组(P<0.05),T等位基因频率高于对照组(P<0.05);在个人过敏史、家族哮喘史临床特征中,哮喘儿童SOCS3基因rs4969169位点基因型分布比较,差异均有统计学意义(P<0.05),且个人过敏史、家族哮喘史儿童SOCS3基因rs4969169位点TT基因型频率分别高于无过敏史和无家族哮喘史儿童(P<0.05);以rs4969169 CC基因型作为参照,携带CT和TT基因型儿童哮喘易感性是携带CC儿童的1.258倍和1.606倍,具有更高的易感性(P<0.05)。结论SOCS3基因rs4969169位点TT基因型和T等位基因可能是儿童哮喘发病的相关优势基因,可能与儿童哮喘易感性有关。Objective To explore the relationship between the cytokine signal transduction factor 3(SOCS3)gene polymorphism and the susceptibility of bronchial asthma in children.Methods 106 children with bronchial asthma received in the outpatient clinic of the hospital from January 2017 to January 2020 were selected as the study group,and 98 healthy children who came to the hospital for physical examination during the same period were selected as the control group.DNA was extracted from two groups of children's peripheral blood,the SOCS3 gene rs4969169 locus genotype were detected,the two groups of children SOCS3 genotype and allele frequency were compared,the relationship between the genotype distribution of SOCS3 gene rs4969169 locus and general information in the research group was analyzed.The relationship between SOCS3 gene rs4969169 locus genotypes and susceptibility to bronchial asthma in children were analyzed.Results The frequency of TT genotype at rs4969169 of SOCS3 gene in the study group was higher than that in the control group(P<0.05),The frequency of T allele was higher than that of the control group(P<0.05).Among the clinical features of personal allergy history and family history of asthma,the genotype distribution of SOCS3 gene rs4969169 in asthmatic children showed statistically significant differences(P<0.05),and the frequency of TT genotype at rs4969169 of SOCS3 gene in children with personal allergy history and family history of asthma were higher than that of children without history of allergy and family history of asthma(P<0.05).Taking the rs4969169 TT genotype as a reference,children with CT and TT genotypes were 1.258 times and 1.606 times more susceptible to asthma than children with CC,and had higher susceptibility(P<0.05).Conclusion The TT genotype and T allele at the rs4969169 locus of SOCS3 gene may be the relevant dominant genes in the onset of childhood asthma,and may be related to the susceptibility of childhood asthma.
