摘 要:目的探究脂质过氧化反应在纳米细菌(NB)损伤HK-2细胞过程中的作用。方法将NB组(NB菌液、胎牛血清、1640培养液+HK-2细胞)、四环素干扰组(四环素、NB菌液、胎牛血清、1640培养液+HK-2细胞)和空白对照组(胎牛血清、1640培养液+HK-2细胞)分别培养6、12、24 h,观察HK-2细胞的形态学变化。用超声粉碎HK-2细胞后检测Na^(+)/K^(+)ATP酶和Ca^(2+)/Mg^(2+)ATP酶活性,并检测培养液中乳酸脱氢酶(LDH)、过氧化氢(H_(2)O_(2))和丙二醛(MDA)的含量。结果HE结果可见,四环素干扰组中HK-2细胞的损伤程度明显低于NB组,干扰组细胞在共培养12、24 h时酶活性高于NB组(P<0.05)。在共培养12、24 h时,NB组培养液中H_(2)O_(2)和MDA的含量显著高于对照组(P<0.05),干扰组显著低于NB组(P<0.05)。结论NB损伤人肾小管上皮细胞HK-2的过程中,引起脂质过氧化反应,并且脂质过氧化反应的程度与人肾小管上皮细胞HK-2损伤程度成正相关。Objective To explore the effects of lipid peroxidation on human renal tubular epithelial HK-2 cells damaged by nanobacteria(NB).Methods HK-2 cells were divided into 3 groups:control group,NB group and tetracycline interference group.Hk-2 cells were co-cultured with each group for 6 h,12 h and 24 h,respectively.Morphological changes of HK-2 cells were observed with a light microscope.The activities of Na^(+)/K^(+)ATpase and Ca^(2+)/Mg^(2+)ATpase were detected after HK-2 cells were crushed with ultrasound.The contents of lactate dehydrogenase(LDH),hydrogen peroxide(H_(2)O_(2))and malondialdehyde(MDA)in the culture medium were determined.Results In NB group,tetracycline interference group and control group,the damage degree of HK-2 cells decreased successively,the enzyme activity increased successively,and the contents of H_(2)O_(2) and MDA decreased successively.Conclusion Lipid peroxidation is induced in the process of NB injury to HK-2 cells,and the degree of lipid peroxidation is positively correlated with the degree of injury.
