摘　　要：研究样本来自黑龙江省绥化市的一个生猪养殖场,分别采集疑似发生猪流感的病猪鼻拭子样本和濒死猪的肺组织样本,处理后接种到10日龄的SPF鸡胚中,分离出一株具有血凝性的病毒。采用血清学、RT-PCR、电镜、生物学特性及理化特性测定等方法对病毒进行鉴定,结果显示分离株的鸡红细胞的凝集价为1:256,只能够中和H1亚型猪流感特异性阳性血清;RT-PCR结果为H1、N1亚型,电镜下的病毒粒子为球形,直径约80 nm,具有囊膜,同时表面有刺突和纤突,对鸡胚的半数感染量(EI D)为10/0.1 mL;该病毒对热、胰蛋白酶、氯仿及乙醚均敏感;从以上病毒特性的研究,可以判定分离得到的病毒为H1N1亚型的猪流感病毒。The samples of this study were collected from a pig farm in Suihua City,Heilongjiang Province,which collected nasal swabs from pigs suspected of swine flu and lung tissue from dying pigs.After treatment,they were inoculated into 10 days SPF chicken embryos,and a virus with hemagglutination was isolated.The virus was identified by serology,RT-PCR,electron microscopy,biological characteristics and physicochemical characteristics.The results showed that the agglutination valence of chicken erythrocytes of the isolated strain was 1:256,which could only neutralize the specificity of H1 subtype swine influenza.Positive serum,RT-PCR results were H1 and N1 subtypes,the virus particles under the electron microscope were spherical,with a diameter of about 80 nm,with a capsule,and there were spikes and fibers on the surface.10/0.1 mL,the virus was sensitive to heat,trypsin,chloroform and ether,from the above study of virus characteristics,it could be determined that the isolated virus was a swine influenza virus of H1N1 subtype.
