丹蒌片改善大鼠急性心肌缺血的作用研究

摘　　要：目的分析丹蒌片改善心肌缺血的保护作用、量效关系及作用机制。方法皮下注射异丙肾上腺素(85 mg/kg)复制心肌缺血模型。实验分为现象实验、量效实验、机制实验3个方面。现象实验大鼠随机分为对照组、异丙肾上腺素组、丹蒌片1000 mg/kg组、美托洛尔组,每组10只,给药2 d;量效实验大鼠随机分为对照组、异丙肾上腺素组、美托洛尔组、丹蒌片5 mg/kg组、丹蒌片50 mg/kg组、丹蒌片100 mg/kg组、丹蒌片500 mg/kg组、丹蒌片5000 mg/kg组,每组10只,给药2 d;机制实验大鼠随机分为异丙肾上腺素组、丹蒌片组、PD98059组、LY294002组、KN93组、L-NAME组,每组10只,给药2 d。通过测定血清肌钙蛋白T(cTnT)和心肌酶5项[天门冬氨酸氨基转移酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)、α-羟丁酸脱氢酶(α-HBDH)]水平,分析丹蒌片改善心肌缺血作用。结果现象实验:与对照组比较,给药后异丙肾上腺素组AST、CK、CK-MB、LDH、α-HBD、cTnT水平均升高(P<0.05);与异丙肾上腺素组比较,丹蒌片1000 mg/kg组和美托洛尔组给药后AST、CK、CK-MB、LDH、α-HBD、cTnT水平均降低(P<0.05)。量效实验:与对照组比较,异丙肾上腺素组给药后AST、CK、CK-MB、LDH、α-HBD、cTnT水平均升高(P<0.05);与异丙肾上腺素组比较,美托洛尔组、丹蒌片5mg/kg组、丹蒌片50mg/kg组、丹蒌片100mg/kg组、丹蒌片500mg/kg组、丹蒌片5000 mg/kg组给药后AST、CK、CK-MB、LDH、α-HBD、cTnT水平均下降(P<0.05),其中,不同丹蒌片给药剂量组AST、CK、CK-MB、LDH、α-HBD、cTnT水平下降幅度不同,且丹蒌片100 mg/kg组各指标下降幅度最大。机制实验:与对照组比较,异丙肾上腺素组给药后AST、CK、CK-MB、LDH、α-HBD、cTnT水平均升高(P<0.05);与异丙肾上腺素组比较,丹蒌片组、PD98059组、LY294002组、L-NAME组、KN93组给药后心肌酶AST、CK、CK-MB、LDH、α-HBD及cTnT水平均下降(P<0.05)。结�Objective To analyze the protective effect,dose-effect relationship and mechanism of Danlou Tablet on myocardial ischemia.Methods Myocardial ischemia model was induced by subcutaneous injection of isoproterenol(ISO,85 mg/kg).The experiments were divided into three aspects:phenomenon experiment,dose-effect experiment and mechanism experiment.In phenomenon experiment,the rats were randomly divided into control group,isoproterenol group,Danlou Tablet 1000 mg/kg group and metoprolol group,with 10 rats in each group,for 2 days.In dose-effect experiment,the rats were randomly divided into control group,isoproterenol group,metoproprol group,Danlou Tablet 5 mg/kg group,Danlou Tablet 50 mg/kg group,Danlou Tablet 100 mg/kg group,Danlou Tablet 500 mg/kg group,and Danlou Tablet 5000 mg/kg group,10 rats in each group,for 2 days.In mechanism experiments,rats were randomly divided into isoproterenol group,Danlou Tablet group,PD98059 group,LY294002 group,KN93 group and L-NAME group,10 rats in each group,for 2 days.The effects of Danlou Tablets on myocardial ischemia were analyzed by measuring serum levels of troponin T(cTnT)and aspartate aminotransferase(AST),creatine kinase(CK),lactate dehydrogenase(LDH),creatine kinase MB isoenzyme(CK-MB),α-hydroxybutyrate dehydrogenase(HBDH).Results In phenomenon experiment,compared with the control group,the levels of AST,CK,CK-MB,LDH,α-HBD,and cTnT in the isoproterenol group were higher than those in the control group(P<0.05);compared with isoproterenol group,the levels of AST,CK,CK-MB,LDH,α-HBD,and cTnT in Danlu Tablet 1000 mg/kg group and metoproprol group were significantly lower(P<0.05).In dose-effect experiment:compared to the control group,the levels of AST,CK,CK-MB,LDH,α-HBD,and cTnT in isoproterenol group was increased(P<0.05);compared with isoproterenol group,the levels of AST,CK,CK-MB,LDH,α-HBD,and cTnT in 1000 mg/kg group were decreased in metoprolol group,Danlou Tablet 5 mg/kg group,Danlou Tablet 50 mg/kg group,Danlou Tablet 100 mg/kg group,Danlou Tablet 500 mg/kg group,and
