摘  要:探究了微生物法合成银纳米团簇的过氧化物模拟酶性质及其应用于血糖浓度检测的可行性。采用催化稳态动力学分析银纳米团簇的过氧化物模拟酶性质,考察pH、温度和银纳米团簇浓度对酶活性的影响,确定其检测H_(2)O_(2)和葡萄糖的线性范围,并应用于小鼠血样中葡萄糖浓度的检测。结果表明,微生物法合成的银纳米团簇具备过氧化物模拟酶性质,在最适条件下,其干重过氧化物模拟酶活性为193.7 U·g^(-1)。其对H_(2)O_(2)和葡萄糖的线性检测范围分别为0.5~4 mmoL·L^(-1)和0.5~3 mmoL·L^(-1)。对小鼠血糖浓度的检测结果与GOPOD氧化酶法试剂盒的结果接近。基于银纳米团簇的过氧化物模拟酶性质,建立了灵敏检测H_(2)O_(2)和葡萄糖浓度的新方法,该方法在血糖检测的实际应用中具有可行性,在分析检测领域中具有良好的发展前景。The properties of peroxidase mimetic enzyme of silver nanoclusters synthesized by microbial method and the feasibility of its application in the detection of blood glucose concentration were explored.The catalytic steady-state kinetics was used to analyze the peroxidease mimetic enzyme properties of silver nanoclusters,and the effects of pH,temperature and silver nanocluster concentration on enzyme activity were investigated,and the linear range for the detection of H_(2)O_(2) and glucose was determined.Then the perxidase mimetic enzyme was also applied to the detection of glucose concentration in mouse blood samples.The results showed that the silver nanoclusters synthesized by the microbial method had the property of peroxidase mimetic enzyme,and the dry weight peroxidase mimetic enzyme activity was 193.7 U·g^(-1) under the optimal condition.The linear detection range for H_(2)O_(2) and glucose was 0.5~4 mmoL·L^(-1) and 0.5~3 mmoL·L^(-1),respectively.The detection result of the mouse blood glucose concentration was close to that of the GOPOD oxidase method kit.Based on the peroxidase mimetic enzyme properties of silver nanoclusters,a new sensitive method for the detection of H_(2)O_(2) and glucose concentration was established,which is feasible in the practical application of blood glucose detection and has a good development prospect in the fields of analysis and detection.
