摘 要:为了解决黑木耳生产过程中菌种退化的问题,提高退化菌种的品质及产量,通过改良母种培养基的方法使菌种复壮,根据菌丝生长速度、密度、生物量、纤维素酶活性及出菇实验后子实体的产量等指标,从3种不同添加物的母种培养基中筛选出最优的母种复壮培养基。结果表明,在PDA培养基中添加稻杆浸出液且浓度为15 g·L^(-1)时,复壮效果最好,经过该培养基复壮后,菌丝密度增大,菌丝生长速度达到4.16 mm·d^(-1),菌丝生物量干重为1.301 g·100 mL^(-1),酶活性为3.86 U·mL^(-1),生物学效率达到82.3%,产量增加了29.5%,菌丝体品质及子实体的产量均有显著提高。选择合适添加物及正确添加量的母种复壮培养基,可以使退化的菌种得到很好的复壮。In order to solve the problem of bacterial degradation in the production process of black fungus and improve the quality and yield of the degraded bacterial species,the method of improving the parent seed medium was used to rejuvenate the bacterial species.According to the indexes of mycelial growth rate,density,biomass,cellulase activity and fruiting body yield after fruiting experiment,the optimal mother seed rejuvenation medium was screened from three different supplemented parent seed media.The results show that the rejuvenation effect is the best when rice straw extract is added to the PDA medium at the concentration of 15 g·L^(-1).After the rejuvenation of the medium,the density of the mycelium increases,and the growth rate of the mycelium reaches 4.16 mm·d^(-1).The biomass dry weight is 1.301 g·100 mL^(-1),the enzyme activity is 3.86 U·mL^(-1),the biological efficiency reaches 82.3%,the yield increases by 29.5%,and the quality of mycelium and the yield of fruiting bodies are significantly improved.The degenerated strains can be rejuvenated well by selecting the appropriate supplements and the correct amount of the parent seed rejuvenation medium.
