摘 要:为探讨鸭感染H6N6亚型禽流感病毒(AIV)后对免疫器官中细胞因子IL-2、IL-10和IFN-α转录水平的影响,建立荧光定量PCR方法对胸腺、脾脏、法氏囊中的IL-2、IL-10和IFN-α3种细胞因子检测,研究非免疫鸭感染H6N6亚型AIV后1、3、5 d各细胞因子在免疫器官中的变化。结果显示,建立的荧光定量PCR方法标准曲线相关系数R~2为0.996~1.000,扩增效率为92.2%~93.6%,相关性较好,熔解曲线整齐单一,变异系数均小于9%,可用于后续试验;鸭在感染H6N6亚型AIV后的一定时间内免疫器官中的IL-2、IL-10和IFN-α呈现不同程度的转录水平变化,以及不同的细胞因子在不同组织之间的转录水平差异较大,其中IL-2在法氏囊中的转录水平比在胸腺和脾脏中变化明显较大;而IFN-α在胸腺和脾脏中的转录水平高于法氏囊;IL-10在免疫器官中转录水平较低。还发现H6N6亚型AIV感染鸭后免疫器官中的IL-2、IL-10、IFN-α转录水平在一定时间内为先上升后下降趋势,并且在感染后5 d降到最低。表明感染H6N6亚型AIV对鸭免疫器官中IL-2、IL-10和IFN-α转录水平有一定的影响。To explore the effect of H6N6 subtype avian influenza virus(AIV)on transcription levels of cytokines IL-2,IL-10and IIFN-αin immune organs of ducks,the IL-2,IL-10and IIFN-αin the thymus,spleen,and bursa of Fabricius were detected based on fluorescence quantitative PCR method established in this experiment,to study the variation of these three cytokines in immune organs after non immune ducks infected with H6N6 subtype avian influenza virus(AIV)within 1,3,5days.The results showed that the standard curve correlation coefficient R2 was between 0.996-1.000,the amplification efficiency reached92.2%-93.6%,the correlation was well related,the melting curve was neat and single,and the coefficient of variation was less than 9%,using this established method,which could be used for follow-up experiment.The IL-2,IL-10and IIFN-αin the immune organs of ducks after infection with the H6N6virus showed varying degrees of transcription level changes,and the transcription levels of different cytokines in different tissues varied greatly.It was proved that the transcription level of IL-2in the bursa of Fabricius varied significantly than in the thymus and spleen;while the transcription levels of IFN-αin thymus and spleen were higher than that in bursa of Fabricius;and that of IL-10was low in immune organs.In addition,it was also found that the transcription levels of IL-2,IL-10and IFN-αin the immune organs of ducks infected with H6N6 subtype AIV generally increased firstly and then decreased within a certain period of time,and decreased to the minimum on the 5th day after infection.The test results confirmed that infection with H6N6 subtype AIV has a certain effect on the transcription levels of IL-2,IL-10and IFN-αin the immune organs of ducks.
