摘  要:为探究国内流行的牛结节性皮肤病病毒(Lumpy skin disease virus,LSDV)野毒株在鼠源BHK-21细胞中的复制生长特性,用间接免疫荧光、蛋白免疫印迹和透射电镜方法,确定了在BHK-21细胞系中能够感染中国流行的LSDV/FJ/CHA/2021毒株。蛋白免疫印迹和病毒生长曲线结果均表明,LSDV野毒株呈现时间依赖性的方式在BHK-21细胞中进行有效复制。电镜结果证明LSDV病毒工厂位于BHK-21细胞质内而非细胞核内。结果表明,LSDV国内流行毒株能够在BHK-21细胞中进行高效复制,为探究LSDV感染的致病机制、动物模型建立及弱毒疫苗的研制奠定重要理论基础。This study aimed to explore the replication and growth characteristics of the domestic epidemic lumpy skin disease virus(LSDV)wild-type strain in hamster origin BHK-21 cells.In this study,the LSDV/FJ/CHA/2021 strain,which is epidemic in China,can infect and replicate in the BHK-21 cell line was identified by indirect immunofluorescence,Western blot,and transmission electron microscopy methods.The Western blot and one-step growth curve results showed that LSDV wild strain could replicate effectively in BHK-21 cells in a time-dependent manner.The results of the transmission electron microscope further proved that the viral factory of LSDV is located in the cytoplasm of BHK-21 rather than in the nucleus.Our results showed that the domestic epidemic strain of LSDV can replicate efficiently in BHK-21 cells,which plays a vital theoretical foundation for further exploring the infection and pathogenesis mechanism of LSDV,establishing animal models,and developing attenuated vaccines.
