摘  要:对患乳房炎牦牛鲜乳的致病菌分离培养、形态学观察、革兰氏染色、生化鉴定、16S rRNA基因序列及遗传进化分析,对所分离菌株进行药敏和致病性试验。结果显示,分离菌株为需氧型革兰氏阳性球菌,其生长特性、菌落形态特征、生化试验结果与葡萄球菌属中的人葡萄球菌基本一致。分离菌对庆大霉素、环丙沙星、利奈唑啉、万古霉素等12种抗菌药物敏感,对克林霉素中度敏感,对红霉素耐药。动物试验中小鼠出现死亡,剖检后发现腹腔积液浑浊,肾脏、肝脏和肺脏均出现不同程度的病变,表明分离菌具有一定毒力。16S rRNA基因序列比对分析和遗传进化分析结果表明该菌株为人葡萄球菌。综上可判定分离菌株为人葡萄球菌,表明人葡萄球菌是西藏牦牛乳房炎的致病原因之一。In order to explore the pathogenic bacteria of yak with mastitis in Tibet,the pathogenic bacteria isolation and culture,morphological observation,Gram staining,biochemical identification,16S rRNA gene sequence identification and genetic evolution analysis were used to study the fresh milk of yak with mastitis.Meanwhile,antimicrobial susceptibility analysis and animal pathogenicity tests were also performed.The results showed that the isolate was aerobic Gram-positive,and the growth characteristics,colony morphology and biochemical identification revealed that the isolate was basically consistent with the Staphylococcus hominis.The antimicrobial susceptibility test showed that the isolate was sensitive to 12 tested antimicrobials,such as gentamicin,ciprofloxacin,linezolid,vancomycin and so on,moderately sensitive to clindamycin,while resistant to erythrocin.The result of pathogenicity test showed that the isolated strain was pathogenic to laboratory mice,the tested mice died after inoculation,the cloudy fluid was found in the abdominal cavity of mice,and varying degrees of pathological lesions were also seen in the kidney,liver and lung.The alignment analysis of 16S rRNA gene sequence and genetic evolution analysis further indicated that the isolated strain was Staphylococcus humanis.In conclusion,our results showed that the isolated strain from the yak with mastitis was Staphylococcus humanis,and it was one of the pathogenic bacteria of yak mastitis in Tibet.
