摘 要:目的:通过动物实验,研究参苓白术散早期干预对过敏性结肠炎(AC)幼鼠肠道炎症反应的影响,探索中医药防治AC的药理机制。方法:150只7 d龄健康雌性BALB/c幼鼠,随机分为空白组、模型组、益生菌组及参苓白术散高、中、低剂量组,益生菌组予口服酪酸梭菌,参苓白术散组予口服高、中、低剂量参苓白术散煎液,空白组及模型组予口服等体积PBS溶液。以噁唑酮诱导制备过敏性结肠炎模型,进行疾病活动指数(DAI)评分,并用HE染色法观察幼鼠结肠组织病理学改变及评分(HPS),ELISA法测定结肠组织白细胞介素4(IL-4)、白细胞介素13(IL-13)、白细胞介素10(IL-10)及干扰素-γ(IFN-γ)含量,Western blot法检测结肠组织早幼粒白血病锌指蛋白(PLZF)的表达水平。结果:灌肠后第1、3天,模型组DAI评分高于空白组,差异具有统计学意义(P<0.05,P<0.01),提示AC模型构建成功;益生菌组及参苓白术散高、中、低剂量组与模型组之间则无统计学差异(P>0.05)。各组间HPS评分结果差异无统计学意义(P>0.05)。参苓白术散高剂量组IL-10、IFN-γ含量均高于模型组,其中IL-4、IL-10、IL-13、IFN-γ的增高有统计学差异(P<0.01,P<0.05),并显著高于益生菌组(均为P<0.01)。各组间PLZF蛋白表达水平结果差异无统计学意义(P>0.05);而益生菌组及参苓白术散高剂量组的PLZF蛋白表达水平与模型组间也无显著差异(P>0.05)。结论:参苓白术散的早期干预,虽未能有效减轻AC幼鼠的过敏性炎症反应,但可显著增加相应抑制性细胞因子IL-10、IFN-γ的表达水平。Objective: This research aims to evaluate the effect of early intervention of Shenlingbaizhu powder on the intestinal inflammation in juvenile mice with allergic colitis(AC), and to explore the pharmacological mechanism of Chinese medicine in preventing and treating AC. Methods: One hundred and fifty healthy female BALB/c juvenile mice aged 7 days were randomly divided into control group, model group, C.butyricum group and Shenlingbaizhu powder groups of high, medium and low dose, respectively. The mice in C.butyricum group were orally given Clostridium butyricum, and those in Shenlingbaizhu powder groups were orally given decoction of Shenlingbaizhu powder of high, medium and low doses, and those in control group and model group were given same volume of PBS orally. Allergic colitis were induced by oxazolone. After induction, disease activity index(DAI) score was measured, and the histopathological changes of mice colon was observed by HE staining method and scored. ELISA was used to detect the contents of interleukin 4(IL-4), interleukin 13(IL-13), interleukin 10(IL-10) and interferon-γ(IFN-γ) in colon tissues. Western blot was used to detect the expression level of promyelocytic leukemia zinc finger protein(PLZF) in colon tissue. Results: On the 1 st and 3 rd day after enema of oxazolone, the DAI score of model group was significantly higher than that of control group(P<0.05, P<0.01), which indicated the success of AC model. There was no significant difference in HPS score among all groups(P>0.05). The histopathological score was not significantly different among all groups(P>0.05). The contents of IL-4, IL-10, IL-13 and IFN-γin Shenlingbaizhu powder group of high dose were higher than those in model group, and the increase of IL-10 and IFN-γwas of statistical significance(P<0.01, P<0.05), which were also higher than those in C.butyricum group(P<0.01).
