摘  要:目的比较神农香菊不同海拔生境的主要生态环境因子差异和基因表达谱差异,研究神农香菊适应不同海拔生境的分子机制。方法利用WheatA获得海拔相关的主要生态因子如温度和降水等并进行差异统计分析,利用转录组测序获得黄龙堰与宋洛两个不同海拔生境的神农香菊样本的基因表达谱,并基于GO、KEGG等数据库进行基因注释、功能富集分析,获取样本间的差异基因及其关联的代谢通路。结果光照和温度是影响神农香菊生长发育的最重要的生态因子。不同海拔种植点样本的差异表达基因主要富集到植物激素信号转导、萜类、苯丙素类、不饱和脂肪酸类等次生代谢生物合成途径。其中,参与萜类生物合成途径的相关基因在不同海拔种植样本间显著上调或下调表达。结论神农香菊可能通过调控萜类关键表达基因如DXS、DXR、HMGCS等的表达适应生境变化。研究结果为深入研究神农香菊不同海拔适应机制及新品种培育提供数据支撑。Objective To study the molecular mechanism of Chrysanthemum indicum var. aromaticum adapting to different altitude habitats by comparing the differences in main ecological environment factors and gene expression profiles in different altitude habitats of C. indicum var. aromaticum.Methods The main ecological factors related to altitude, such as temperature and precipitation, were obtained by using the WheatA, and the difference statistical analysis was carried out. The gene expression profiles of C. indicum var. aromaticum samples from two different altitude habitats, Huanglongyan and Songluo, were obtained by transcriptome sequencing. Based on GO, KEGG and other databases for gene annotation and functional enrichment analysis obtained differential genes between samples and their associated metabolic pathways.Results Light and temperature were the most important ecological factors affecting the growth and development of C. indicum var. aromaticum. The differential expressed genes of the samples from the two planting areas were significantly enriched in the plant hormone signal transduction and the biosynthetic pathways of terpenoids, phenylpropanoids, unsaturated fatty acids and other secondary metabolites biosynthesis pathways, among which genes involved in terpenoid biosynthesis were significantly up-regulated or down-regulated in samples grown at different altitudes.Conclusion The results indicate that C. indicum var. aromaticum may adapt to habitat changes by regulating the expression of key terpenoid expression genes such as DXS, DXR, and HMGCS. The research results provide data support for in-depth research on the high-altitude adaptation mechanism and the cultivation of new varieties of C. indicum var. aromaticum.
