摘  要:为探讨刺五加皂苷B对小鼠下丘脑抗冷应激的调控作用,以小鼠下丘脑为模型,应用实时荧光定量PCR和Western-blot方法,测定相关基因Hsp70、CRH、ACTH、TRH mRNA和蛋白表达量。结果显示,冷应激条件下,4 mg/kg刺五加皂苷B对小鼠下丘脑抗冷应激相关因子和激素具有上调的作用,能够显著提高Hsp70、ACTH、TRH、CRH蛋白和基因的相对表达量。说明刺五加皂苷B对冷应激小鼠下丘脑具有保护作用,此保护作用是通过诱导激活SAM系统、HPA轴和HPT轴,从而增加下丘脑中ACTH、TRH、CRH等抗应激激素含量,并且使得应激蛋白Hsp70表达增强,提高冷应激小鼠抗冷应激能力。To investigate the regulation of Acanthopanax senticosus saponin B on hypothalamus cold stress in mice,real-time fluorescent quantitative PCR and Western-blot were used to determine the expression of Hsp70,CRH,ACTH,TRH genes and protein in the hypothalamus of mice.The results showed that 4 mg/kg saponin B could up-regulate the anti-cold stress related factors and hormones in the hypothalamus of mice under cold stress conditions,and significantly increase the relative expression of Hsp70,ACTH,TRH,CRH proteins and genes.It is concluded that Acanthopanax senticosus saponin B has protective effect on hypothalamus of mice under cold stress.The protective effect of Acanthopanax senticosus saponin B is to increase the content of ACTH,TRH and CRH in hypothalamus by inducing the activation of SAM system,HPA axis and HPT axis,and to enhance the expression of stress protein Hsp70,so as to enhance the ability of cold stress mice to resist cold stress.
