摘  要:为了获得高质量的口蹄疫疫苗,对悬浮培养技术和抗原的浓缩纯化技术进行改进优化,细胞悬浮培养方式由批式培养改为连续流悬浮培养,增加离心纯化次数和超滤系统,最终可缩短48 h以上的细胞培养时间,获得高纯度和高含量的浓缩抗原,所制备的疫苗理化性状、安全性、有效性、保存期等质量指标均获得显著改善。这一技术的改进为生产高质量的口蹄疫疫苗提供了技术支持,并可实现规模化生产。The optimized cell suspension culture technology and antigen concentration and purification technology were used in the production of foot-and-mouth disease vaccine to shorten the cell culture time,improve the production efficiency and the stability of antigen.The quality of foot-and-mouth disease vaccine was improved obviously,and the character,antigen content,emulsified particle size and immune protection of the vaccine remained unchanged after storage for 30 months.The improved continuous cell flow culture technology and antigen concentration and purification technology can prepare more stable,higher purity,higher content of vaccine antigen and safe,effective,stable and long shelf life of foot-and-mouth disease vaccine products,and can realize large-scale production.
