摘 要:研究白族惯用药鞍叶羊蹄甲水提物对小鼠的急性毒性,确定鞍叶羊蹄甲的半数致死量(LD_(50)),考察鞍叶羊蹄甲的安全性。LD_(50)实验采用寇氏法给药,观察小鼠7 d内临床症状,解剖观察其实质脏器病理变化;最大给药量(MLD_(50))实验将小鼠随机分为3组,口服灌胃给药,连续观察14 d内小鼠的毒性反应和死亡情况,并对实验小鼠进行血常规和血液生化指标检测及组织病理学观察。LD_(50)实验小鼠无死亡,实质脏器无眼观病理变化;MLD_(50)实验结果显示LD_(50)>150 g/kg,属实际无毒型,第14天组织病理学观察发现小鼠肝脏、肾脏均有不同程度损伤,考虑为超剂量用药所致。结果提示,鞍叶羊蹄甲具有良好的安全性。The objective was to study the acute toxicity of water extract from Bauhinia brachycarpa to mouse,and investigate the safety of Bauhinia brachycarpa according to the median lethal dose ( LD_(50)) .In the LD_(50)experiment,Kou’s method was used to administer the drug.The clinical symptoms of the mouse was observed within 7 days,and the organ morphology was observed after 7 days.In the maximum dose ( MLD_(50)) experiment,the mice were randomly divided into 3 groups and given by gavage administration.Continuously observe the toxicity and death of mouse within 14 days,and perform blood gas analysis and pathological tissue examination on experimental mouse.The results showed that no mouse died and no abnormal tissue examination in LD_(50)experiment;MLD_(50) experiment showed that LD_(50)> 150 g·kg,which was actually non-toxic,varying degrees of damage was observed in the mouse’s liver and kidney on the 14th day of administration,which might be caused by overdose and course of treatment.The water extract of Bauhinia brachycarpa has good safety,and the study can provide a theoretical basis for the clinical application of Bauhinia brachycarpa.
