摘 要:通过GC-MS分析艳山姜精油挥发性组分,并利用人体肝癌细胞HepG2系及CCK法、流式细胞法、细胞增殖相关基因表达分析法,研究艳山姜精油抑制肿瘤细胞增殖的作用。结果表明:艳山姜精油挥发组分主要包含烯烃类和醇类物质,分别占挥发组分的63.88%、19.52%;在精油浓度为200μg/mL时,处理48h后对细胞增殖的抑制率达到56.8%;采用200μg/mL的艳山姜精油处理HepG2细胞24h,细胞凋亡率提高了2.5倍,同时,促进细胞凋亡的基因Caspase-3、Caspase-9、p53表达量提升至对照的2.57、1.74、1.50倍,而促进细胞增殖的PCNA基因表达量降低到0.45倍。艳山姜精油能较好地抑制癌细胞HepG2的增殖,为利用艳山姜开发功能食品提供了支撑。The volatile components of Alpinia zerumbet(Pers.) Burtt. et Smith essential oil(AEO) were analyzed by GC-MS,and the inhibitory effect of AEO on tumor cell proliferation was studied using human hepatoma cell HepG2, with CCK method,flow cytometry and cell proliferation related gene expression was also analyzed. The results showed that the volatile components of AEO mainly contains olefins and alcohols, accounting for 63.88% and 19.52% of the volatile components respectively.When the concentration of essential oil was 200 μg·mL–1, the inhibition rate of cell proliferation reached 56.8% after 48 hours of treatment;The apoptosis rate of HepG2 cells treated with 200 μg·mL–1 essential oil increased by 2.5 times. At the same time,the expression of Caspase-3, caspase-9 and p53 increased by 2.57, 1.74 and 1.50 times, while the expression of PCNA gene promoting cell proliferation decreased to 0.45 times. This study showed that AEO can better inhibit the proliferation of cancer cell HepG2, which provides support for Alpinia zerumbet to develop functional food.
