摘 要:在草莓脱毒苗组织培养过程中,继代和生根是重要的步骤。不同的增殖系数和生根状态对后续脱毒苗的成活和产量起关键作用。本实验在草莓脱毒苗继代培养过程中,于培养基中添加不同浓度比例的六苄基腺嘌呤(6-BA)、吲哚丁酸(IBA)和马铃薯汁,促进继代和生根草莓脱毒苗生长。结果显示,草莓脱毒苗继代增殖效果最佳的培养基是MS+0.8 mg/L 6-BA+0.2 mg/L IBA+100 g/L马铃薯汁,增殖系数为4.17。生根培养过程中,通过对比MS、1/2 MS及是否添加萘乙酸(NAA)等不同处理,得出1/2 MS+0.1 mg/L NAA效果最佳,生根系数最高且根系短粗,有利于移栽驯化。Subculture and rooting are quite important steps during virus-free strawberry seedlings tissue culture.Different multiplication coefficient and rooting situation play a key role in surviving rate and mass production of virus-free seedlings.Different concentration proportion of 6-BA,IBA and potato juice had been added to the subculture culture medium,in order to improve the growth rate of subculture and rooting of virus-free strawberry seedlings.Results showed that the optimal culture medium was MS+0.8 mg/L 6-BA+0.2 mg/L IBA+100 g/L potato juice by choosing the higher multiplication coefficient.NAA was added or not in the MS or 1/2 MS medium during rooting,data showed that the optimal culture medium was 1/2 MS+0.1 mg/L NAA,by analyzing the rooting rate and the rooting situation,and root system was short and thick,which was beneficial to the following transplanting domestication.
