摘 要:研究黄樟鞘花Macrosolen cochinchinensis(Lour.)Van Tiegh.茎叶的化学成分及其抗肿瘤活性。采用硅胶、大孔树脂、聚酰胺、MCI Gel、ODS、Sephadex LH-20柱色谱及高效制备液相色谱法对黄樟鞘花乙醇提取物进行分离纯化,运用现代波谱技术鉴定了14个化合物,分别为β-香树酯醇乙酸酯(1)、β-香树酯醇(2)、羽扇豆醇(3)、环桉树醇(4)、菜油甾醇(5)、β-谷甾醇(6)、槲皮素(7)、槲皮苷(8)、没食子酸(9)、芦丁(10)、鞣花酸(11)、短叶苏木酚(12)、鞣花酸-3,3′-二甲醚-4′-O-β-D-木糖苷(13)、鞣花酸-3,3′-二甲醚-4-O-β-D-木糖苷(14),14个化合物均为首次从黄樟鞘花中分离得到。以A549肺癌细胞为研究对象,采用CCK-8细胞增殖检测试剂盒测定化合物1~14对A549肺癌细胞的抑制效果。利用流式细胞仪分析化合物14对A549肺癌细胞周期分布和凋亡水平变化的影响。研究发现除化合物1、2和4外,其余化合物对A549细胞均有不同程度的抑制作用(IC=9.3~42.2μg/mL),以化合物14作用最强,其细胞周期被阻滞在G1期,细胞凋亡率显著增加。研究表明化合物3和化合物5~14可能是鞘花主要抗肿瘤活性成分,化合物14能抑制A549肺癌细胞增殖,其机制可能是通过阻滞细胞周期从而诱导细胞凋亡。To study the chemical constituents and antitumor activities in the stems and leaves of Macrosolen cochinchinensis(Lour.)Van Tiegh.Silica gel,macroporous resin,polyamide,MCI Gel,ODS,Sephadex LH-20 column chromatography and preparative high performance liquid chromatography were used to separate and purify ethanol extract from M.cochinchinensis,and modern spectroscopic techniques were used to identify 14 compounds,namelyβ-amyrin acetate(1),β-amyrin(2),lupeol(3),cycloeucalenol(4),campesterol(5),β-sitosterol(6),quercetin(7),quercitrin(8),gallic acid(9),rutin(10),ellagic acid(11),brevifolin(12),ellagic acid-4′-O-β-D-xylopyranoside-3,3′-dimethylether(13)and ellagic acid-4′-O-β-D-xylopyranoside-3,3′-dimethylether(14).All 14 compounds were isolated from M.cochinchinensis for the first time.Taking A549 lung cancer cells as the research object,the inhibitory effect of compounds 1-14 on A549 lung cancer cells was determined by CCK-8 cell proliferation test kit.The effects of compound 14 on cell cycle distribution and apoptosis level change of A549 lung cancer cells were analyzed by flow cytometry.It was found that all compounds except compounds 1,2 and 4 had different inhibitory effects on A549 cells(IC=9.3-42.2μg/mL),and compound 14 had the strongest effect,and its cell cycle was blocked in G1 phase,and the apoptosis rate increased significantly.Compound 3 and compounds 5-14 may be the main antitumor active components of M.cochinchinensis.Compound 14 can inhibit the proliferation of A549 lung cancer cells,and its mechanism may be to induce apoptosis by blocking cell cycle.
