摘 要:目的:探究熄风化湿方对腹泻型肠易激综合征(diarrheal irritable bowel syndrome,IBS-D)内脏高敏感性大鼠的作用及其机制。方法:从40只SPF级雄性Wistar大鼠中随机选取10只作为空白组,其余30只大鼠采用“母婴分离+乙酸刺激”法建立IBS-D模型。将建模成功的IBS-D大鼠随机分成模型组、低剂量熄风化湿方组(10.9 g·kg^(-1))和高剂量熄风化湿方组(43.6 g·kg^(-1)),每组10只,低、高剂量熄风化湿方组大鼠灌胃给予10 mL·kg^(-1)的熄风化湿方溶液,空白组和模型组大鼠灌胃给予等体积蒸馏水,每天1次,给药2周。收集各组大鼠粪便,检测粪便含水率;检测大鼠腹部抬起和背部拱起所需注水量评价大鼠内脏敏感性;HE染色检测大鼠结肠组织病理变化;ELISA法检测血清生长激素释放肽(growth hormone releasing peptide,Ghrelin)、胆囊收缩素(cholecystokinin,CCK)、白细胞介素-1β(interleukin-1β,IL-1β)及IL-2的水平;免疫荧光染色检测肠黏膜γδT细胞数量;Western Blotting检测酪氨酸蛋白激酶(tyrosine-protein kinase,Jak)、信号转导激活转录因子(signal transducer activated transcription factor,Stat)蛋白表达水平。结果:与空白组比较,模型组大鼠腹部抬起和背部拱起所需注水量明显降低(P<0.05);粪便含水率及血清Ghrelin、CCK、IL-1β水平显著上升(P<0.05),IL-2水平显著降低(P<0.05);肠黏膜γδT细胞数量及结肠组织Jak、Stat蛋白表达水平显著升高(P<0.05)。与模型组比较,低、高剂量熄风化湿方组大鼠腹部抬起和背部拱起所需注水量明显升高(P<0.05);粪便含水率及血清Ghrelin、CCK、IL-1β水平显著降低(P<0.05),IL-2水平显著上升(P<0.05);肠黏膜γδT细胞数量及结肠组织Jak、Stat蛋白表达水平显著降低(P<0.05)。HE染色发现,模型组大鼠结肠组织固有层有明显中性粒细胞浸润,间质水肿严重,腺体排列紊乱,边界不清晰,黏膜下层可见血管增生;低、高剂量熄风化湿方组大鼠结�Objective:To explore the effect and mechanism of Xifeng Huashi Recipe on visceral hypersensitivity rats with diarrhea-type irritable bowel syndrome(IBS-D).Methods:10 of 40 SPF male Wistar rats were randomly selected as blank group,and the remaining 30 rats were established in the IBS-D model by the"mother-infant separation+acetic acid stimulation"method.The successfully modeled IBS-D rats were randomly divided into a model group,a low-dose Xifeng Huashi Recipe group(10.9 g·kg^(-1)),and a high-dose Xifeng Huashi Recipe group(43.6 g·kg^(-1)),with 10 rats in each group.Rats in low-dose and high-dose Xifeng Huashi Recipe groups were given 10 mL·kg^(-1) of Xifeng Huashi Recipe solution by gavage,while rats in the blank group and model group were given an equal volume of distilled water by gavage,once a day for 2 weeks.The feces of the rats in each group were collected,and the water content of the feces was detected.Measure the water injection volume required for lifting the abdomen and arching the back of the rat to evaluate the visceral sensitivity of the rat.HE staining was used to detect the pathological changes of rat colon tissue.Serum levels of growth hormone releasing peptide(Ghrelin),cholecystokinin(CCK),interleukin-1β(IL-1β),and IL-2 were detected by ELISA.The number ofγδT cells in intestinal mucosa was detected by immunofluorescence staining.Western Blotting was used to detect the protein expression levels of the tyrosine-protein kinase(Jak)and signal transducer activated transcription factor(Stat).Results:Compared with the blank group,the water injection volume required for abdominal lifting and back arching of rats in the model group was significantly reduced(P<0.05).The water content of feces and the levels of serum Ghrelin,CCK,and IL-1βwere significantly increased(P<0.05),while the level of IL-2 was significantly decreased(P<0.05).The number of intestinal mucosalγδT cells and the expression levels of Jak and Stat proteins in colon tissue were significantly decreased(P<0.05).HE staining showed t
