仙乐雄胶囊对环磷酰胺诱导的小鼠生精功能损伤的保护作用

摘  要:目的:探讨仙乐雄胶囊对环磷酰胺(CP)诱导小鼠生精功能损伤的保护作用。方法:将60只雄性ICR小鼠随机分为6组,依次为正常对照组、CP模型组、仙乐雄低剂量组、仙乐雄高剂量组、生精胶囊组、复方玄驹胶囊组。除正常对照组给予同等剂量的溶媒外,其余各组给予相应药物灌胃30 d,自第25天开始模型组及各给药组以CP(60 mg/kg,腹腔注射)进行造模,持续5 d,正常对照组腹腔注射等剂量生理盐水。造模结束后次日测量各小鼠相关脏器指数、精液浓度、精子畸形率,HE染色评估睾丸与附睾尾组织,检测外周血清睾酮(T)水平以及睾丸氧化应激相关酶活力。结果:与其他组相比,CP模型组小鼠性器官质量和指数、精液浓度、外周血清T下降(P<0.05),精子畸形率增加(P<0.05),睾丸及附睾组织出现病理性损伤;高剂量仙乐雄胶囊(600 mg/kg)可改善上述指标(P<0.05),且精子计数、外周血清T水平等指标的改善优于生精胶囊或复方玄驹胶囊(P<0.05);仙乐雄胶囊改善了CP诱导的小鼠睾丸组织超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽还原酶(GR)以及谷胱甘肽过氧化物酶(GSH-px)活性,下调了MDA(P<0.05)。结论:仙乐雄胶囊可通过增强CP诱导的睾丸组织各类抗氧化酶活性,进而改善睾丸组织氧化应激损伤,具有重要的临床价值。Objective:To investigate the protective effect of Xianlexiong capsule on the injury of spermatogeny in mice induced by cyclophosphamide(CP).Methods:Sixty male ICR mice were randomized to normal control group, CP model group, low dose Xianlexiong group, high dose Xianlexiong group, Shengjing capsule group and compound Xuanju capsule group.Mice in each group, except normal control group, received intragastric administration with corresponding drugs for 30 days.By day 25,model group and all drug administration groups were given CP(60 mg/kg, intraperitoneal injection) for modeling for 5 days, and mice in normal control group were injected intraperitoneally with equal dose of normal saline.The indexes of related sex organs, semen concentration and sperm malformation rate of each mouse were measured the next day after modeling.Testicular and caudal epididymis tissues were observed via HE staining.Testosterone(T) level in peripheral blood and testicular oxidative stress related enzyme activity were also measured.Results:The sex organ weight or index, semen concentration and T in peripheral blood of mice in CP model group were significantly decreased(P<0.05),and the rate of sperm malformation was markedly increased(P<0.05),with notable pathological damage of the testicular and epididymis tissues.However, high dose Xianlexiong capsule(600 mg/kg) could significantly improve the above indexes(P<0.05),and the improvement of sperm count and peripheral blood T level were significantly better in mice treated with high dose Xianlexiong than in those managed by either Shengjing capsule or compound Xuanju capsule(P<0.05).In addition, Xianlexiong capsule led to improved activities of OXIDE dismutase(SOD),malondialdehyde(MDA),glutathione reductase(GR) and glutathione peroxidase(GSH-Px) in CP induced testicular tissue of mice, and down-regulated level of MDA(P<0.05).Conclusion:Xianlexiong capsule has important clinical value, because it can enhance the activity of various antioxidant enzymes in the testis tissues induced by CP,thus i
