摘 要:目的:变应性鼻炎(AR)是常见的过敏性疾病之一,传统中药黄芩的活性成分之一黄芩苷元,具有良好的抗过敏作用,本文探讨黄芩苷元制剂对AR豚鼠的症状评分、血清中白细胞介素4及免疫球蛋白E含量的影响,为黄芩苷元的临床运用提供实验依据。方法:40只成年Hartley豚鼠,雌雄各半,随机分成模型组(0 mL),低剂量组(雾化量0.2 m L/min),中剂量组(0.5 m L/min)以及高剂量组(1 mL/min),每组10只。各组采用卵清蛋白雾化吸入+冷水浸足方法建立变应性鼻炎模型。建模后除了模型组外,其余各组豚鼠分别使用不同剂量的黄芩苷元鼻喷雾剂超声雾化吸入治疗10 min,每日2次,连用2周。通过比较豚鼠症状评分及血清白细胞介素4 (IL-4)含量、免疫球蛋白E (IgE)蛋白表达情况评估黄芩苷元鼻喷雾剂的疗效。结果:建模成功后所有大鼠的症状评分均在5分以上,经黄芩苷元鼻喷雾剂雾化治疗2周后,模型组症状评分仍最高,黄芩苷元鼻喷雾剂干预后评分有所下降,差异有统计学意义(F=4.237,P<0.05)。中、高剂量组血清IL-4水平较模型组显著降低,差异有统计学意义(t=4.258,8.364,P<0.05)。黄芩苷元鼻喷雾剂干预组IgE表达较模型组降低,差异有统计学意义(F=4.759,P<0.05)。结论:黄芩苷元能够有效减轻豚鼠AR导致的过敏反应,为临床治疗此类疾病提供了新的思路。Objective: Allergic rhinitis is one of the common allergic diseases. Baicalin, one of the active components of traditional Chinese medicine Scutellaria baicalensis, has a good anti-allergic effect. This paper intends to investigate the effect of baicalin preparation on the symptom score of allergic rhinitis(AR) guinea pigs and the content of interleukin-4 and immunoglobulin E in serum, so as to provide experimental basis for the clinical application of baicalin. Methods: 40 adult Hartley guinea pigs, half male and half female, were randomly divided into model group(0 m L), low-dose group(aerosolized volume of 0.2 m L/min), medium-dose group(0.5 mL/min) and high-dose group(1 mL/min), with 10 in each group. Allergic rhinitis models were established by the method of ovalbumin inhalation and foot immersion in cold water in each group. After modeling, except for the model group, guinea pigs in other groups were treated with different doses of baicalin nasal spray by ultrasonic atomization inhalation for 10 min, twice a day, for 2 weeks. The efficacy of baicalin nasal spray was evaluated by comparing guinea pig symptom scores, serum interleukin-4(IL-4) content and immunoglobulin E(IgE) protein expression. Results: After successful modeling, the symptom scores of all rats were above 5 points. After 2 weeks of atomization treatment with baicalin nasal spray, the symptom score of the model group was still the highest, and the score decreased after the intervention of baicalin nasal spray, and the difference was statistically significant(F=4.237, P<0.05). The serum IL-4 levels in the middle and high dose groups were significantly lower than those in the model group, and the difference was statistically significant(t=4.258, 8.364, P<0.05). The expression of IgE in the baicalin nasal spray intervention group was lower than that in the model group, and the difference was statistically significant(F=4.759, P<0.05).
