摘  要:喜树(Camptotheca acuminata)能够合成具有抗肿瘤作用的喜树碱(CPT)与10-羟喜树碱(10-HCPT),其积累具有组织特异性,但对于同一组织不同发育阶段的积累特性尚不清楚。利用高效液相色谱和实时荧光定量PCR分析喜树幼叶不同部位(叶片、叶基、叶柄)CPT和10-HCPT积累量以及关键酶基因相对表达量。结果发现,幼叶中CPT的积累量在叶片中最多,其次是叶基、叶柄;10-HCPT的积累量在叶柄中最多,其次是叶基、叶片;合成途径关键酶基因CaG8O、CaCYC1、Ca7-DLS、Ca7-DLGT、CaTDC 1以及CaSTR的表达均具有相似的趋势,且与CPT的积累量呈负相关,但与10-HCPT的积累量呈正相关。Camptothecin(CPT)and 10-hydroxycamptothecin(10-HCPT)with anti-tumor activity can be biosynthesized in Camptotheca acuminata,and their accumulations are tissue specific.However,the accumulation characteristics of the same tissue at different developmental stages are still unclear.The accumulations of CPT and 10-HCPT and relative expression of key enzyme genes in different parts of young leaves(leaf,leaf base,petioles)were analyzed by HPLC and qRT-PCR.The results show that the highest accumulation of CPT in young leaves is leaf,followed by leaf base and petiole.The highest accumulation of 10-HCPT is in petioles,followed by leaf base and the least in leaf.The expression pattern of the key enzyme genes(CaG8O,CaCYC1,Ca7-DLS,Ca7-DLGT,CaTDC 1 and CaSTR)also show similar tendency,which is negatively correlated with the accumulation of CPT,positively correlated with the accumulation of 10-HCPT in these three parts of young leaves.
