摘  要:目的:探讨大黄䗪虫丸(DHZCP)对肝癌大鼠糖脂代谢与免疫功能的影响。方法:24只SD大鼠随机分为空白组,模型组,DHZCP低、高剂量组,每组6只。模型组和两给药组大鼠按50 mg/kg剂量腹腔注射二乙基亚硝胺,每周2次,4周之后改为每周1次,持续至第12周。空白组腹腔注射0.9%氯化钠溶液,注射体积为2 mL/kg。DHZCP低、高剂量组大鼠自造模第3周起灌胃给药,DHZCP用量根据临床用药剂量折算,低、高剂量分别为0.5、1.0 g/kg,每日1次,持续灌胃至第21周。检测大黄䗪虫丸对肝癌大鼠肝脏和血清中糖脂代谢关键酶及葡萄糖、游离脂肪酸含量的影响;对CD_(8)^(+)T细胞组织分布、细胞因子TNF-α的影响;对CD_(4)^(+)T细胞亚群Th1、Th2、Th17、Treg细胞比例及相应细胞因子的影响。结果:与空白组比较,模型组大鼠血清AFP-L、APT含量显著升高(P<0.01)。与模型组比较,DHZCP低、高剂量组大鼠糖脂代谢关键酶HK、FAS、ACC、CPT-1及游离脂肪酸的含量,Th2、Treg细胞比例及细胞因子水平显著降低(P<0.01,P<0.05);葡萄糖含量,CD_(8)^(+)T细胞在肝组织中的比例及细胞因子TNF-α水平,Th1、Th17细胞比例及细胞因子水平显著升高(P<0.01,P<0.05)。结论:DHZCP可调节肝癌的异常糖脂代谢,减少肝脏的糖酵解、脂肪酸合成与氧化,增加CD_(8)^(+)T细胞在肝组织中的比例,调节CD_(8)^(+)T细胞与CD_(4)^(+)T细胞亚群之间的平衡,协同发挥抗肿瘤作用。Objective:To explore the regulating effect of Dahuang Zhechong Pills (DHZCP) on glucose and lipid metabolism and immune function in rats with liver cancer.Methods:Twenty-four SD rats were randomly divided into blank group,model group,low and high dose DHZCP groups with 6 rats in each group.Rats in the model group and the two administration groups were intraperitoneally injected with diethylnirtosamine at a dose of 50 mg/kg,twice a week,and then changed to once a week after 4 weeks,and continued to the 12th week.The blank group was intraperitoneally injected with 0.9% sodium chloride solution,and the injection volume was 2 mL/kg.The rats in the low and high dose groups of DHZCP were given intragastric administration from the third week of model establishment.The dosage of DHZCP was converted according to the clinical dose,and the low and high doses were 0.5 and 1.0 g/kg,respectively,once a day,and continued to be administered by intragastric administration until week 21.The effect of DHZCP on the key enzymes of glucose and lipid metabolism in liver and serum of liver cancer rats and the content of glucose and nonestesterified fatty acid;the effect on CD_(8)^(+)T cell tissue distribution and cytokine TNF-α;and the effect on CD_(4)^(+)T cell subsets including Th1,Th2,Th17,Treg cell ratio and the influence of corresponding cytokines were investigated.Results:Compared with the blank group,the serum AFP-L and APT levels of the model group were increased (P<0.01).Compared with the model group,the contents of key enzymes of glucose and lipid metabolism HK,FAS,ACC,CPT-1 and nonestesterified fatty acid,the proportion of Th2 and Treg cells and the levels of cytokines in the low and high dose DHZCP groups were significantly decreased (P<0.01,P<0.05);the glucose content,the proportion of CD_(8)^(+)T cells in liver tissue and the level of cytokine TNF-α,the proportion of Th1 and Th17 cells and the level of cytokines were significantly increased (P<0.01,P<0.05).Conclusion:DHZCP can regulate abnormal glucose and lipid metaboli
