摘 要:目的:探讨青赤散对溃疡性结肠炎(UC)小鼠肠道菌群及Treg/Th17免疫平衡的作用机制。方法:将48只C57BL/6J小鼠按随机数表法随机分为空白组,模型组,美沙拉秦组,青赤散低、中、高剂量组,每组8只。采用葡聚糖硫酸钠(DSS)构建小鼠UC模型,监测小鼠体质量,进行DAI评分,测量结肠的长度和重量;HE染色观察青赤散对UC小鼠肠黏膜炎症影响;肠道菌群分析和短链脂肪酸含量测定监测肠道菌群中微生物多样性、丰度以及短链脂肪酸的含量;流式细胞术测定小鼠肠系膜淋巴结中Treg/Th17的细胞比例。结果:与空白组比较,模型组小鼠体质量降低,DAI评分升高,结肠的长度和重量降低,肠道菌群的多样性和丰度减少,短链脂肪酸含量降低,Treg/Th17的细胞比例失衡(P<0.05,P<0.01);与模型组比较,各给药组小鼠体质量上升,DAI评分降低,结肠的长度和重量增加,肠道菌群的多样性和丰度增多,短链脂肪酸含量增多,Treg/Th17的细胞比例恢复正常水平(P<0.05,P<0.01)。结论:青赤散可以通过调节肠道菌群和Treg/Th17免疫平衡治疗小鼠UC。Objective:To investigate the mechanism of Qingchi Powder on intestinal flora and Treg/Th17 immune balance in mice with ulcerative colitis(UC).Methods:A total of 48 C57BL/6J mice were randomly divided into blank group,model group,mesalazine group,Qingchi Powder low,medium and high dose groups,with 8 mice in each group.The UC model of mice was established by dextran sodium sulfate(DSS).The body weight,DAI score,colon length and weight of mice were monitored.HE staining was used to observe the effect of Qingchi Powder on intestinal mucosal inflammation in mice with UC.The intestinal flora analysis and short-chain fatty acid content were used to monitor the diversity and abundance of microorganisms in the intestinal flora and the content of short-chain fatty acids.The proportion of Treg/Th17 cells in mouse mesenteric lymph nodes was determined by flow cytometry.Results:Compared with the blank group,the body weight of mice in the model group was decreased,the enteritis score was increased,the length and weight of colon were decreased,the diversity and abundance of intestinal flora were decreased,the content of short-chain fatty acids was decreased,and the proportion of Treg/Th17 cells was unbalanced(P<0.05,P<0.01).Compared with model group,different doses of Qingchi Powder treatment groups of body weights were gained in mice,the enteritis scores were reduced,the length and weight of the colon were increased,the diversity and abundance of the intestinal flora were increased,short chain fatty acid contents were increased,the proportion of Th17/Treg cells was gradually returned to normal level(P<0.05,P<0.01).Conclusion:Qingchi Powder can treat UC in mice by regulating the intestinal flora and Treg/Th17 immune balance.
