摘  要:目的:筛选新五汁饮对虚热证小鼠药效最优的组方。方法:70只雄性昆明小鼠适应性喂养7d后随机分为正常组,模型组,六味地黄丸组,新五汁饮Ⅰ、Ⅱ、Ⅲ、Ⅳ组,每组10只。甲状腺片灌胃造虚热证模型。连续2周灌胃给药。造模及给药期间检测小鼠耳温、体质量变化。末次给药后检测小鼠血常规,摘取小鼠甲状腺、肺组织,HE染色后观察组织变化情况,并进行免疫组化检测甲状腺组织、肺组织NF-κB p65、p-NF-κB p65、I-κBβ蛋白表达变化,比较不同组方的新五汁饮对虚热证药效的影响。结果:第14天,与正常组比较,模型组小鼠体质量显著降低(P<0.01),耳温显著升高(P<0.01);与模型组比较,各治疗组小鼠体质量显著升高(P<0.01),耳温显著降低(P<0.01)。与正常组比较,模型组小鼠白细胞计数(WBC)及中性粒细胞百分比(NEUT%)显著升高(P<0.01);与模型组比较,六味地黄丸组及新五汁饮Ⅱ、Ⅲ、Ⅳ组WBC显著降低(P<0.01),六味地黄丸组、新五汁饮Ⅳ组NEUT%显著降低(P<0.01)。六味地黄丸组及新五汁饮Ⅳ组甲状腺状态接近正常组;六味地黄丸组及新五汁饮Ⅲ、Ⅳ组肺组织状态接近正常组。与正常组比较,模型组甲状腺NF-κB p65、p-NF-κB p65、I-κBβ蛋白表达水平显著升高(P<0.05,P<0.01);与模型组比较,新五汁饮Ⅰ、Ⅱ、Ⅲ、Ⅳ组甲状腺组织NF-κB p65蛋白表达水平显著降低(P<0.05),六味地黄丸组及新五汁饮Ⅰ、Ⅱ、Ⅳ组甲状腺组织p-NF-κB p65蛋白表达水平显著降低(P<0.05),六味地黄丸组,新五汁饮Ⅰ、Ⅲ、Ⅳ组I-κBβ蛋白表达水平显著降低(P<0.05)。与正常组比较,模型组肺组织NF-κB p65、p-NF-κB p65、I-κBβ蛋白表达水平显著升高(P<0.05);与模型组比较,新五汁饮Ⅳ组肺组织NF-κB p65、p-NF-κB p65、I-κBβ蛋白表达水平显著降低(P<0.05)。结论:新五汁饮Ⅳ组对虚热证模型小鼠药效最优。Objective:To screen the optimal pharmacological efficacy of New Five-juice Decoction on mice with deficiency heat evidence model.Methods:Seventy male Kunming mice were randomly divided into normal group,model group,Liuwei Dihuang Pills group,New Five-juice Decoction Ⅰ,Ⅱ,Ⅲ and Ⅳ groups after 7 d of adaptive feeding,and 10 mice in each group.Gastric gavage was administered continuously for 2 weeks.The changes of ear temperature and body mass of mice were detected during the modeling and drug administration.After the last administration,the blood routine of mice was checked.The thyroid and lung tissues of mice were extracted,and the tissue changes were observed after HE staining.Moreover,immunohistochemistry was performed to detect the changes of NF-κB p65,p-NF-κB p65 and I-κBβ protein expression in thyroid tissue and lung tissue,comparing the effects of different groups of New Five-juice Decoction on the medicinal efficacy of deficiency heat evidence.Results:On day 14,the body mass of mice in the model group was significantly lower(P<0.01) and the ear temperature was significantly higher(P<0.01) compared with the normal group.In comparison with the model group,the body mass of mice in each treatment group was dramatically higher(P<0.01) and the ear temperature was remarkably lower(P<0.01).The white blood cell count(WBC) and neutrophil percentage(NEUT%) of mice in the model group were dramatically higher than those in the normal group(P<0.01);when compared with the model group,the WBC was lowered seriously(P<0.01) in Liuwei Dihuang Pills group,New Five-juice Decoction Group Ⅱ,Ⅲ,and Ⅳ;the NEUT% was reduced considerably(P<0.01) in Liuwei Dihuang Pills group and the new five-juice drinking Ⅳ group.The thyroid status of Liuwei Dihuang Pills group and New Five Juice Drink Ⅳ group was close to the normal group;the lung tissue status of Liuwei Dihuang Pills group,New Five Juice Drink Ⅲ group and New Five Juice Drink IV group was close to the normal group.In comparison with the normal group,the expre
