摘  要:目的:分析芩百清肺浓缩丸治疗感染后咳嗽(PIC)的效应机制。方法:烟熏、辣椒素雾化吸入联合脂多糖腹腔注射进行PIC造模。以阿斯美为对照,0.14 g/m芩百水溶液以1 mL/100 g体质量灌胃给药10 d。辣椒素雾化吸入诱咳。ELISA法测定血清P物质(SP),支气管肺泡灌洗液(BALF)中SP、瞬时感受器电位阳离子通道香草酸受体1(TRPV1)含量。RT-PCR法测定BALF和肺组织中TRPV1、M2受体mRNA水平。Western Blot法检测肺组织TRPV1、M2受体蛋白表达水平。结果:与模型组比较,芩百组与阿斯美组血清和BALF中SP含量显著减少(P<0.05),BALF中TRPV1含量显著减少(P<0.05),BALF中TRPV1 mRNA表达水平显著降低(P<0.05),肺组织中TRPV1蛋白表达显著降低(P<0.05),BALF和肺组织M2受体mRNA表达水平、肺组织M2受体蛋白表达显著增加(P<0.05)。与阿斯美组比较,芩百组BALF中SP含量显著减少(P<0.05),肺组织TRPV1 mRNA表达水平显著增加(P<0.05),肺组织M2受体mRNA及蛋白表达显著增加(P<0.05),BALF中M2受体mRNA表达显著增加(P<0.05)。结论:TRPV1激活在PIC发病中作用更大。芩百清肺浓缩丸能够降低PIC的SP水平、降低TRPV1基因表达、修复和上调M2受体。Objective:To analyze the mechanism of Qinbai Qingfei Condensed Pills(Qinbai) on post-infectious cough(PIC).Methods:PIC guinea pigs were modeled by smoking,capsaicin aerosol inhalation combined with lipopolysaccharide(LPS) intraperitoneal injection.0.14 g/mL Qinbai aqueous solution was administered by gavage at 1 mL/100 g body mass for 10 days with the Asmine compared.The contents of substance P(SP) and transient receptor potential vanilloid 1(TRPV1) were determined by ELISA,the mRNA levels of TRPV1 and M2 receptors were determined by RT-PCR,the expression of TRPV1 and M2 receptor protein in lung tissue was detected by Western Blot.Results:Compared with the model group,SP and TRPV1 content were significantly decreased in Qinbai group and Asimei group,the expression level of TRPV1 mRNA in BALF,the expression level of TRPV1 mRNA in lung tissue and the expression level of TRPV1 protein in Asimei group were significantly decreased,both mRNA and protein expression of M2 receptor were significantly increased(P<0.05).Compared Asimei group,the amount of SP in BALF decreased,the expression of M2 receptor protein of Qinbai group was significantly increased(P<0.05).Conclusion:The activation of TRPV1 may play a greater role in PIC.Qinbai Qingfei Condensed Pills can reduce the levels of SP,reduce the expression of TRPV1 mRNA,repair and raise the level of the damaged M2 receptor in PIC.
