摘 要:目的:通过观测孤束核(NTS)胃饥饿素(Ghrelin)、胃饥饿素受体(GHS-R)及瘦素受体(Ob-R)的表达变化,探讨逍遥散影响慢性束缚应激(CIS)肝郁脾虚大鼠体质量与摄食量变化的机制。方法:60只SD大鼠随机分为正常组、模型组、氟西汀组和逍遥散组,每组15只。每天3 h连续21 d CIS方法建立动物模型,同时灌胃给予相应药物。观察各组大鼠体质量、摄食和旷场实验行为学指标变化;间苯三酚法测定血清D-木糖含量;RT-qPCR、免疫组化和Western Blot法分别检测各组大鼠NTS中Ghrelin、GHS-R、Ob-R mRNA或蛋白表达。结果:与正常组比较,模型组大鼠体质量降低,摄食量下降,旷场实验总移动距离、站立和修饰次数显著减少,血清D-木糖含量显著下降(P<0.05);NTS中Ghrelin、GHS-R阳性细胞表达数和总积分光密度显著减少(P<0.05),Ob-R阳性细胞表达数和总积分光密度显著升高(P<0.05);GHS-R mRNA和蛋白表达显著减少(P<0.05),Ob-R mRNA和蛋白表达显著升高(P<0.05)。与模型组比较,逍遥散组大鼠体质量、摄食量及旷场实验总移动距离、站立次数和血清D-木糖含量显著升高(P<0.05);NTS中Ghrelin、GHS-R表达显著升高(P<0.05),Ob-R表达显著降低(P<0.05)。结论:逍遥散可通过调节CIS大鼠NTS中Ghrelin、GHS-R和Ob-R的表达变化,发挥调节食欲、抗应激作用。Objective:Through the observation of the expression changes of Ghrelin,growth hormone secretagogue receptor(GHS-R),and leptin receptor(Ob-R) in the nucleus of solitary tract(NTS),to explore the mechanism of Xiaoyaosan on the changes in food intake and body weight of chronic immobilization stress(CIS) rats with liver stagnation and spleen deficiency.Methods:Sixty SD rats were randomly divided into normal group,model group,fluoxetine group and Xiaoyaosan group,with 15 rats in each group.A 21-day chronic immobilization stress model was established for 3 hours every day,and corresponding drugs were given,and the changes in body weight,food intake and open field experiment of rats in each group were observed;phloroglucinol method was used to measure serum D-xylose content;RT-qPCR,immunohistochemistry and western bloting were used to detect the gene or protein expression of Ghrelin,GHS-R,and Ob-R in the rats’ NTS of each group.Results:Compared with the normal group,the body weight and food intake of the model group were obviously decreased,the total moving distance,standing and modification times in the open field experiment were significantly reduced,the serum D-xylose content was apparently declined(P<0.05);the positive cells and integrated optical densities of Ghrelin and GHS-R in NTS were dropped evidently(P<0.05),while expression of Ob-R were increased prominently(P<0.05);the gene and protein expression of GHS-R in NTS were lessened notably(P<0.05),and Ob-R expression promoted greatly(P<0.05).Compared with the model group,the body weight,food intake,total moving distance,standing times and serum D-xylose content of the Xiaoyaosan group were improve markedly(P<0.05);the expression of Ghrelin and GHS-R in NTS were increased prominently,while Ob-R expression lessened notably(P<0.05).Conclusion:Xiaoyaosan can regulate appetite and anti-stress by adjusting the expression of Ghrelin,GHS-R and OB-R in NTS of CIS rats.
