泄泻湿热证及寒湿证患者舌苔与舌面菌群相关性研究

摘　　要：目的:通过分析泄泻湿热证、寒湿证患者及健康人群舌面菌群特征、舌苔客观化数据及其相关性,为舌诊研究的宏观与微观有机结合探索新思路。方法:采集泄泻湿热证、寒湿证患者及健康志愿者舌苔样本、舌图像,提取舌苔DNA进行高通量测序,获得样本物种信息和舌图像信息,分析上述结果及两者相关性。结果:拟杆菌门、普雷沃氏菌属在泄泻患者舌面丰度显著高于健康组(P<0.05),健康组舌面变形菌门、奈瑟氏菌属、梭杆菌属和嗜血杆菌属丰度显著高于泄泻患者(P<0.05),卟啉单胞菌属和假单胞菌属在泄泻寒湿证患者舌面丰度显著低于泄泻湿热患者和健康组(P<0.05);泄泻患者舌苔面积占比显著增高、L值显著降低,湿热证泄泻患者舌苔G值、B值、S值、a值均显著升高(P<0.05);链球菌属与舌苔R值、G值、B值均呈正相关(P<0.05),奈瑟氏菌属、嗜血杆菌属和拟普雷沃氏菌属与舌苔H值呈负相关(P<0.05)。结论:舌面菌群可随疾病和证型发生改变,且由病证和舌面环境共同作用;链球菌属和拟普雷沃氏菌属可能与黄苔的形成具有相关性,卟啉单胞菌属和假单胞菌属的减少可能是白腻苔的特征性表现。Objective:To explore new ideas for the organic combination of macroscopic and microscopic research on tongue diagnosis by analyzing the characteristics of tongue flora,objective data of tongue coating and they correlation between the two aspects in patients with diarrhea of damp-heat syndrome,cold-damp syndrome and healthy people.Methods:Collect tongue coating samples and tongue images from patients with damp-heat type and cold-damp type diarrhea and healthy volunteers,extract tongue coating DNA for high-throughput sequencing,obtain sample species information and tongue image information.Analyze the above results and the correlation between the two results.Results:The abundance of Bacteroidetes and Prevotella on the tongue surface of diarrhea patients was significantly higher than that of the healthy group(P<0.05),the abundance of Proteobacteria,Neisseria,Fusobacterium and Haemophilus on the tongue surface of the healthy group was significantly higher than that of the diarrhea patients(P<0.05),while the abundance of Porphyromonas and Pseudomonas on the tongue surface of patients with cold-dampness syndrome diarrhea were significantly lower than those in patients with damp-heat syndrome diarrhea and healthy group(P<0.05).The G value,B value,S value,and a value of the tongue coating in patients with damp-heat syndrome diarrhea are significantly increased(P<0.05),while the L value is significantly reduced and the area of the tongue coating is significantly increased in patients with diarrhea.The color of the tongue coating is correlated with a variety of tongue flora.Among them,Streptococcus is positively correlated with the R value,G value,and B value(P<0.05),Neisseria,Haemophilus and Alloprevotella were negatively correlated with the H value(P<0.05).Conclusion:Tongue flora can change with disease and syndrome types,and is affected by disease syndrome and tongue surface environment.Streptococcus and Prevotella may be related to the formation of yellow coating.The reduction of Porphyromonas and Pseudomonas may be ch
