摘 要:目的:观察安宫降压丸对自发性高血压大鼠(SHR)的降压作用,并对降压机制进行初步探索。方法:SPF级健康雄性SHR随机分为模型组,安宫降压丸高、中、低剂量组和卡托普利组,每组12只,另取同周龄WKY大鼠12只作为正常对照组,各组均连续给药6周。采用无创血压测试仪尾套法测量大鼠尾动脉收缩压(SBP)和舒张压(DBP);采用ELISA法测定血清肾素(REN)、血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)和一氧化氮(NO)水平;采用免疫组化法检测胸主动脉组织中内皮型一氧化氮合酶(eNOS)的表达水平。结果:给药后,与模型组比较,安宫降压丸各剂量组大鼠SBP显著下降(P<0.01);安宫降压丸高剂量组大鼠血清中的AngⅡ、REN和ET-1水平均显著降低(P<0.01,P<0.05),NO水平显著升高(P<0.05);安宫降压丸高、低剂量组大鼠胸主动脉组织中eNOS的表达水平显著增加(P<0.05,P<0.01)。结论:安宫降压丸可通过调节SHR的肾素-血管紧张素-醛固酮系统、改善血管内皮功能等途径发挥降压作用。Objective:To observe the antihypertensive effect of Angong Jiangya Pills on spontaneously hypertensive rats(SHR) and explore its mechanism.Methods:SPF healthy male SHR rats were randomly divided into model group,Angong Jiangya Pills high-dose,medium-dose,low-dose groups and Captopril group,with 12 rats in each group.Another 12 WKY rats of the same strain and age were taken as normal control group,and each group was treated for 6 weeks.The systolic(SBP) and diastolic(DBP) blood pressure of rat tail artery were measured by non-invasive method.Serum renin(REN),angiotensin Ⅱ(AngⅡ),endothelin-1(ET-1) and nitric oxide(NO) were measured by ELISA.The expression of endothelial nitric oxide synthase(eNOS) in thoracic aorta was detected by immunohistochemistry.Results:After drug intervention,compared with model group,the SBP of rats in each dose group of Angong Jiangya Pills significantly decreased(P<0.01).The levels of AngⅡ,REN and ET-1 of Angong Jiangya Pills high-dose group were significantly decreased(P<0.01,P<0.05),and the level of NO was significantly increased(P<0.05).The expression level of eNOS in thoracic aorta of Angong Jiangya Pills high-dose and low-dose groups were significantly increased(P<0.05,P<0.01).Conclusion:Angong Jiangya Pills can reduce blood pressure by regulating REN Angiotensin Aldosterone system and improving vascular endothelial function in SHR rats.
