摘 要:SOC1是控制开花的关键因子,对柑橘的花发育过程起重要作用。以赣南早脐橙为试材,提取成熟叶片总RNA合成cDNA,RT-PCR扩增脐橙CsSOC1a基因CDS全长,对其进行生物信息学分析和花发育不同阶段的表达模式分析。结果表明:脐橙CsSOC1a基因的CDS全长为663 bp,编码220个氨基酸,分子式为C_(1080)H_(1791)N_(331)O_(343)S_(9),分子量大小为25.19 kD,为亲水碱性蛋白;CsSOC1a编码的蛋白质含有MADS-box、K-box和SOC1-motif保守结构域,属于MADS超家族的SOC1/T3亚家族;进化树聚集成3个簇,脐橙SOC1a基因和其他柑橘SOC1基因聚在第2簇。顺势作用元件分析表明CsSOC1a含有光响应元件、植物激素响应元件、低温应答元件等调控元件。相对表达分析发现CsSOC1a主要积累在赣南早脐橙的成熟叶片中,在脐橙开花前和低温处理条件下CsSOC1a相对表达量均显著上调。这些结果表明CsSOC1a基因能够响应低温信号,对赣南早脐橙的成花诱导起促进作用。SOC1 is the key factor controlling flowering and plays an important role in citrus flower development.In this experiment,’Gannanzao’navel orange was used as the test material to extract the total RNA of mature leaves to synthesize c DNA,and the full-length CDS of navel orange SOC1a gene was amplified by RT-PCR.The bioinformatics characteristics and expression pattern at different stages of flower development were analyzed.The results showed that the full-length CDS of CsSOC1a was 663 bp and encoded 220 amino acids.The molecular formula of CsSOC1a gene protein was C_(1080)H_(1791)N_(331)O_(343)S_(9),and its molecular weight was 25.19 k D,which was a hydrophilic alkaline protein.The CsSOC1a had MADS-box,K-box,and SOC1-motif conserved structural domains,and belonged to the SOC1/T3 subfamily of the MADS superfamily.The evolutionary tree was grouped into three subgroups,the CsSOC1a was located in the second subgroup.Cis-acting elements analysis showed that CsSOC1a contained basic promoters and other functional elements such as light response elements,plant hormones response elements,low-temperature response elements and so on.CsSOC1a was mainly accumulated in the mature leaves of’Gannanzao’navel orange.The relative expression level of CsSOC1a was significantly up-regulated before flowering and under low temperature treatment.These results indicated that CsSOC1a gene could respond to low temperature signal and promote floral induction of’Gannanzao’navel orange。
