摘 要:DNA指纹图谱对种质资源鉴定具有重要的指导意义。本研究利用17对多态性SSR引物对29个西兰花品种进行分析,采用荧光M13毛细管电泳技术进行多态检测,共检测到64个多态位点,每对引物的多态位点数达到3.8。标记SSR位点的基因多样性指数变化范围为0.425~0.732,平均值为0.632,多态性信息量(PIC)的变化范围为0.332~0.666,平均值为0.540。29份西兰花品种的聚类分析结构显示,其遗传距离在0.042~0.400之间。根据17对SSR引物在29个品种中扩增的特异带型组合,采用引物-带型组合法构建了29个西兰花品种的指纹图谱,4对SSR特征引物(C2-6,C1-2,C9-29和C9-27)组合可区分所有实验品种。29个西兰花品种的SSR指纹图谱均具有唯一性,每一条指纹信息代表一个独立品种,可为品种鉴定提供依据。DNA fingerprinting is of great significance to the identification of germplasm resources.In this study,17 pairs of polymorphic SSR primers were used to analyze 29 broccoli cultivars hybrids,and the polymorphism was detected by fluorescence M13 capillary electrophoresis.The results showed that total of 64 alleles were amplified with the set of primers,with an average of 3.8.The range of gene diversity and polymorphism information content(PIC)at 17 SSR loci of 29 hybrids were 0.425~0.732 and 0.332~0.666,with an average of 0.632 and 0.540,respectively.The genetic distance among hybrids ranged from 0.042 to 0.400,on which a dendrogram tree showing the relationship among the hybrids was constructed.Among the 17 SSR loci,four primer pairs with high polymorphism(C2-6,C1-2,C9-29 and C9-27)were identified as the specific primers for distinguishing all the 29broccoli hybrids,resulting in the fingerprints for these tested hybrids.These findings data should provide valuable information in hybrid identification in broccoli.
