摘 要:为了考察枫杨提取物(PHHE)对大鼠关节软骨破坏的生物医学影响。通过前交叉韧带横断和内侧半月板切除术(ACLMT)诱导了大鼠创伤后骨关节炎(PTOA)模型。建模成功2周后,将大鼠随机分为ACLMT组(n=10)和PSE组(n=10)。PSE组大鼠每天按照100 mg/kg的剂量(2 mL)腹腔注射PSE溶液,ACLMT组腹腔注射等体积生理盐水(2 mL)。共治疗4周。通过酶联免疫吸附剂法(ELISA)测定大鼠血清中TNF-α和IL-1β的水平。通过苏木精和伊红(HE)以及番红O/固绿染色对大鼠关节组织进行病理学检查。通过免疫组织化学染色及Western blot检查关节软骨组织中Collagen Ⅱ和MMP13的表达。研究结果表明,H&E染色和番红O/固绿染色显示PSE明显减轻了大鼠膝关节的病理变化及软骨破坏。与ACLMT组相比(10.54±0.67,P<0.05),PSE组的Mankin评分显著降低(6.32±0.41,P<0.05)。与ACLMT组相比,PSE组大鼠血清TNF-α和IL-1β水平显著降低(P<0.05)。免疫组织化学染色和Western blot结果显示,与ACLMT组相比,PSE组大鼠关节软骨中Collagen II的蛋白表达水平显著升高,而MMP13显著降低(P<0.05)。上述结果说明,PSE可有效抑制前交叉韧带和半月板损伤大鼠的关节软骨破坏,其机制可能与抑制炎症反应、上调Collagen II的表达并下调MMP13的表达有关。To investigate the effect of Pterocarya stenoptera extract(PSE)on the destruction of articular cartilage in rats with anterior cruciate ligament and meniscus injury.A rat model of post-traumatic osteoarthritis(PTOA)was induced by anterior cruciate ligament transection and medial meniscus resection(ACLMT).Two weeks after the successful modeling,the rats were randomly divided into ACLMT group(n=10)and PSE group(n=10).Rats in the PSE group were intraperitoneally injected with PSE solution at a dose of 100 mg/kg(2 mL),and the ACLMT group was intraperitoneally injected with an equal volume of normal saline(2 mL).A total of 4 weeks of treatment.The levels of TNF-α and IL-1β in rat serum were determined by enzyme-linked immunosorbent method(ELISA).Pathological examination of rat joint tissues was performed by hematoxylin and eosin(HE)and Safranin O/Fast Green staining.The expression of Collagen Ⅱ and MMP13 in articular cartilage tissue was examined by immunohistochemical staining and Western blot.H&E staining and Safranin O/Fast Green staining showed that PSE significantly reduced the pathological changes and cartilage destruction of rat knee joints.Compared with the ACLMT group(10.54±0.67,P<0.05),the Mankin score of the PSE group was significantly lower(6.32±0.41,P<0.05).Compared with the ACLMT group,the levels of TNF-αand IL-1βin the serum of rats in the PSE group were significantly decreased(P<0.05).The results of immunohistochemical staining and Western blot showed that compared with the ACLMT group,the protein expression levels of Collagen II in the articular cartilage of rats in the PSE group increased significantly,while the MMP13 decreased significantly(P<0.05).The above results indicate that PSE can effectively inhibit the destruction of articular cartilage in rats with anterior cruciate ligament and meniscus injury.The mechanism may be related to the inhibition of inflammation,up-regulation of Collagen II expression and down-regulation of MMP13 expression.
