摘 要:目的 探讨病毒miRNA(sv40-miR-S1-5p、ebv-miR-BART8-3p、hsv1-miR-H6-5p、hsv2-miR-H10)与脑室周围白质软化症(PVL)的关系。方法 将2013年1月至2021年8月于南方医科大学珠江医院就诊的7例早产儿脑室周围白质软化症患儿作为试验组,5例健康早产儿为对照组,应用miRNA基因芯片检测两组患儿外周血清中miRNA的表达,筛选出显著差异表达的4个病毒miRNA。本研究应用靶基因预测软件、GO分析、Pathway通路预测对4个病毒miRNA进行生物学分析;应用实时荧光定量PCR(qRT-PCR)对4个病毒miRNA进一步验证其差异表达。结果 4个病毒miRNA采用miRanda及targetscan数据库预测得到靶基因,进行GO分析显示靶基因与炎症、免疫、神经发育等密切相关;经Pathway通路预测其生物学信息通路与炎症、免疫密切相关,其中靶基因IL1R、TLR6、IFN-αβR等位于MAPK和Toll受体信号通路上游的主干位置;采用q RT-PCR对病毒miRNA进行验证,sv40-miR-S1-5p、hsv1-miR-H6-5p、hsv2-miR-H10共3个病毒miRNA的相对表达量与对照组比较,差异有统计学意义(P <0.05),与既往研究miRNA基因芯片检测结果一致。结论 部分病毒miRNA在PVL早产儿外周血清中的表达存在显著差异,其靶基因涉及感染、炎症、免疫等,提示病毒miRNA可能通过作用于其炎症相关靶基因调节机体炎症反应参与PVL疾病的发生。Objective To investigate the relationship between viral miRNA(sv40-miR-S1-5p, ebv-miR-BART8-3p, hsv1-miR-H6-5p, hsv2-miR-H10) and periventricular leukomalacia(PVL). Methods In the preliminary study, 7 premature infants with PVL treated in Zhujiang Hospital from January 2013 to August 2021 were included in the test group, and 5 healthy premature infants were included in the control group. miRNA Microarray was used to detect the expression of miRNA in the peripheral serum of infants in the two groups, in order to screen four viral miRNAs with significant differential expression. In this study, target gene prediction software, GO analysis and Pathway prediction were used to conduct biological analysis for four viral miRNAs;qRT-PCR was used to further verify the differential expression of the four viral miRNAs. Results The target genes of four virus miRNA were predicted by miRanda and targetscan databases, and GO analysis showed that the target genes were closely related to inflammation, immunity, neurodevelopment, etc. The Pathway prediction results showed that the biological information pathways of these target genes were closely related to inflammation and immunity, and the target genes IL1R, TLR6, IFN-αβR and so on were located in the trunk in the upstream of MAPK and Toll receptor signaling pathway;the virus miRNAs were verified by qRT-PCR, and the relative expression levels of the three viral miRNAs sv40-miR-S1-5p, hsv1-miR-H6-5p and hsv2-miR-H10 were significantly different from those of the control group(P < 0.05), which was consistent with the results of miRNA Microarray in previous studies. Conclusion There are significant differences in the expression of some viral miRNAs in the peripheral serum of premature infants with PVL, and their target genes are involved in infection, inflammation, immunity, etc., suggesting that viral miRNAs may participate in the occurrence of PVL disease by regulating the body’s inflammatory response by acting on their inflammation related target genes.
