摘 要:单克隆抗体因高特异性、低副作用等优点,被广泛应用于肿瘤、自身免疫性疾病和传染性疾病的治疗,是当下的研究热点。免疫球蛋白G(IgG)是目前单克隆抗体药物的主要类型,其与可结晶(Fc)段受体的相互作用与多种生物学功能相关,分析IgG与Fc段受体的相互作用是单克隆抗体药物质量控制的重要组成部分。表面等离子共振(SPR)技术是一种新型光学分析技术,可以实时、无标记检测分子间相互作用。近年来,SPR技术被广泛应用于IgG与Fc段受体结合活性的检测。本文综述了基于SPR技术检测IgG与Fcγ受体(FcγRs)、新生儿Fc受体(FcRn)和补体1q(C1q)结合活性的实验方法以及不同方法结果之间的差异和可能引起这些差异的原因。Monoclonal antibodies,due to their advantages of high specificity and low side effects,are widely used in the treatment of tumors,autoimmune diseases and infectious diseases and are currently a research hotspot.Immunoglobulin G(IgG)is the main type of monoclonal antibody drugs at present.Its interaction with crystallizable fragment(Fc)engaging molecules is related to various biological functions.Analysis of the interaction between IgG and Fc engaging molecules is the important part of the quality control of drugs.Surface plasmon resonance(SPR)technology is a novel optical analysis technique that enables real-time,label-free detection of intermolecular interactions.In recent years,SPR technology has been widely used to detect the binding activity of IgG to Fc engaging molecules.This article reviews the experimental methods for detecting the binding activity of IgG to Fcγreceptors(FcγRs),neonatal Fc receptors(FcRn)and complement 1q(C1q)based on SPR technology and summarizes the differences between the results of different methods and the possible reasons for these differences.
