摘  要:目的观察电针对坐骨神经慢性压迫损伤(CCI)大鼠模型脊髓G蛋白偶联受体激酶2(GRK2)及炎症因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)的影响,探讨其缓解CCI疼痛的作用机制。方法将25只大鼠随机分为对照组、模型1组、模型2组、导气电针组和电针组5组,每组5只。造模后3 d对导气电针组和电针组大鼠干预,共干预14次。干预4 d后处死模型1组,末次治疗后处死其余4组。于造模前、治疗前、治疗4 d、治疗11 d和末次治疗后测定各组大鼠机械缩足痛阈(MWT),采用RT-PCR法测定各组大鼠脊髓GRK2 mRNA表达,采用Western blot法测定大鼠脊髓GRK2及IL-1β、TNF-α、IL-10蛋白表达。结果治疗4 d后,与对照组比较,模型1组、模型2组MWT显著下降(P<0.05);与模型2组比较,导气电针组MWT显著上调(P<0.05)。治疗11 d后,与模型2组比较,导气电针组、电针组MWT显著提高(P<0.01)。末次治疗后,与模型2组比较,导气电针组、电针组MWT显著上调(P<0.01);与电针组比较,导气电针组MWT显著上调(P<0.01)。与对照组比较,模型1组和模型2组脊髓GRK2 mRNA和蛋白表达显著降低(P<0.05),脊髓TNF-α、IL-1β蛋白表达显著上升(P<0.05),脊髓IL-10蛋白表达显著下降(P<0.05)。与模型2组比较,导气电针组及电针组脊髓GRK2 mRNA和蛋白表达显著上升(P<0.05)。与模型2组比较,导气电针组及电针组脊髓TNF-α蛋白表达显著下降(P<0.01);与电针组比较,导气电针组脊髓TNF-α蛋白表达显著下降(P<0.01)。与模型2组比较,导气电针组脊髓IL-1β蛋白表达显著下降(P<0.01);与电针组比较,导气电针组脊髓IL-1β蛋白表达显著下降(P<0.05)。与模型2组比较,导气电针组脊髓IL-10蛋白表达显著上升(P<0.01);与电针组比较,导气电针组脊髓IL-10蛋白表达显著上调(P<0.05)。结论电针治疗可以有效降低CCI大鼠的疼痛痛敏并缓解炎症持续趋势,其机制可能与其调节脊髓GRKObjective To observe the impact of electroacupuncture(EA)on G protein-coupled receptor kinase2(GRK2)and inflammatory factors interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and interleukin-10(IL-10)in the spinal cord of rats with chronic constriction injury(CCI)of the sciatic nerve and discuss the mechanism of EA in releasing CCI-induced pain.Method Twenty-five rats were randomized into a control group,model group 1,model group 2,Qi-conduction EA group,and EA group,with 5 rats in each group.Rats in the Qi-conduction EA group and EA group received a total of 14 times of interventions after 3-day modeling.Rats in model group 1 were sacrificed after 4-day intervention,and rats in the other four groups were sacrificed after the last intervention.Before modeling,before treatment,and at 4-day,11-day,and last treatments,the mechanical withdrawal threshold(MWT)was measured,the spinal cord GRK2 mRNA expression was determined using the reverse transcription-polymerase chain reaction(RT-PCR)method.The protein expression levels of GRK2,IL-1β,TNF-α,and IL-10 were detected using the Western blot method.Result After 4-day intervention,the MWT notably dropped in model groups 1 and 2 compared with the control group(P<0.05),and the MWT increased significantly in the Qi-conduction EA group compared with model group 2(P<0.05).After 11-day treatment,the MWT went up significantly in the Qi-conduction EA and EA groups compared with model group 2(P<0.01).After the last treatment,the MWT elevated markedly in the Qi-conduction EA and EA groups compared with model group 2(P<0.01),and the MWT was significantly higher in the Qi-conduction EA group than in the EA group(P<0.01).The GRK2 mRNA and protein expression levels in the spinal cord decreased markedly,the protein expression level of TNF-αand IL-1βin the spinal cord increased markedly,and the protein expression level of IL-10 in the spinal cord decreased markedly in model groups 1 and 2 compared with the control group(P<0.05).The GRK2 mRNA and protein expression levels in th
