摘  要:目的分析江苏省2011-2019年风疹病毒(Rubella virus,RV)流行株基因型和基因亚型。方法对2011-2019年江苏省风疹报告病例采集咽拭子标本,进行RV分离、E1基因核苷酸序列扩增和测定,与世界卫生组织各基因型RV参考株构建亲缘关系树,鉴定RV基因型和基因亚型,分析氨基酸变异。结果从江苏省2011-2019年风疹病例中共分离到19株RV,其中1E基因型13株,包括1E-L1基因亚型7株、1E-L2基因亚型6株;2B基因型6株,均为2B-L1基因亚型。与BRD-II疫苗株相比,1E基因型主要出现A333T(2019年)、L338F(2011-2013年)和L338S(2019年)变异;2B基因型主要出现A333V(2011-2015年)变异。结论江苏省2011-2019年RV流行株出现2个基因亚型传播链更替,即2015年2B-L1取代1E-L1和2019年1E-L2取代2B-L1;需进一步加强RV分子病毒学监测。Objective To determine rubella virus(RV)genotypes and sub-genotypes circulating in Jiangsu province during 2011-2019.Methods We collected throat swab samples from reported rubella cases in Jiangsu during 2011-2019 for RV isolation and E1 gene nucleotide sequence amplification and detection.We constructed phylogenetic trees between the isolated strains and World Health Organization reference strains to identify RV genotypes and sub-genotypes and their amino acid variations.Results A total of 19 RV strains were isolated from rubella cases in Jiangsu during 2011-2019;13 strains were 1 E genotype,including 71 E-L1 sub-genotype and 61 E-L2 sub-genotype;6 strains were 2 B genotype,all 2 B-L1 sub-genotype.Compared with the BRD-II vaccine strain,the 1 E genotype strains had main mutations of A333 T in 2019,L338 F in 2011-2013,and L338 S in 2019;the 2 B genotype strains had main mutations of A333 V in 2011-2015.Conclusions Two sub-genotype RV strains alternately circulated in Jiangsu during 2011-2019;2 B-L1 substituted for 1 E-L1 in 2015 and 1 E-L2 substituted for 2 B-L1 in 2019.We should strengthen RV molecular virology monitoring.
