扇贝裙边糖胺聚糖体外抗HSV-2病毒的实验研究

摘　　要：目的探讨扇贝裙边糖胺聚糖(SS-GAG)对单纯疱疹病毒2型(HSV-2)的体外抗病毒作用。方法观察病毒对宿主细胞(Vero细胞)的形态影响和毒性反应。采用噻唑蓝法检测SS-GAG对Vero细胞的毒性作用以及不同浓度SS-GAG对HSV-2的直接灭活、抑制复制活性及综合作用等。结果SS-GAG在实验浓度范围内对Vero细胞均无毒性。与病毒对照组相比,SS-GAG各浓度组(50、100、200mg/L)能有效保护经HSV-2感染的Vero细胞,使细胞活性增强(F=20.061、39.947,P<0.01),并减弱HSV-2导致的病变效应,抑制病毒的复制,但是SS-GAG对HSV-2没有直接灭活作用。结论SS-GAG在体外实验中显示出保护宿主细胞及抵抗HSV-2感染的活性作用,提示其在抗HSV-2活性方面具有进一步研究的潜力。Objective To investigate the antiviral effect of glycosaminoglycan from scallop skirt(SS-GAG)against herpes simplex virus type 2(HSV-2)in vitro.Methods The influence of HSV-2 on the morphology of host cells(Vero cells)and the toxicity of HSV-2 on cells were observed.MTT assay was used to observe the toxicity of SS-GAG on Vero cells and the effect of different concentrations of SS-GAG on HSV-2 in terms of direct inactivation,inhibition of replication activity,and comprehensive effect.Results SS-GAG had no toxicity to Vero cells within the experimental concentration range.Compared with the virus control group,the SS-GAG treatment groups(50,100,and 200 mg/L)showed effective protection of Vero cells infected with HSV-2 and significantly improved their viability(F=20.061,39.947;P<0.01),and meanwhile,SS-GAG treatment reduced the cytopathic effect induced by HSV-2 and inhibited virus replication,while SS-GAG had no direct inactivation effect on HSV-2.Conclusion SS-GAG has the activity of protecting host cells and defending against HSV-2 infection in vitro,suggesting that further studies are needed to investigate its anti-HSV-2 effect.
