经典藏药佐太对15种人源肿瘤细胞系的体外增殖抑制研究

摘　　要：为了探索经典藏药佐太对人源肿瘤细胞系的体外抑制作用,文章选用11种临床常见癌症(肝癌、乳腺癌、胃癌、肠癌、卵巢癌、宫颈癌、肺癌、胶质母细胞瘤、肾癌、皮肤癌、白血病)15种人源肿瘤细胞系,在体外测定培养基稀释后的佐太5种前处理液(DMSO悬液、未过膜人工胃液浸提液、过膜人工胃液浸提液、未过膜人工肠液浸提液和过膜人工肠液浸提液)对肿瘤细胞的增殖抑制率,计算半数抑制浓度(IC_(50))。实验发现培养基稀释的佐太DMSO悬液对肝癌细胞(BEL-7402和SMMC-7721)、乳腺癌细胞(MDA-MB-231和MCF-7)、胃癌细胞(MKN-45和SGC-7901)、结肠癌细胞HCT 116、卵巢癌细胞(HO-8910和SK-OV-3)、宫颈癌细胞Hela、肺癌细胞A549、脑星型胶质母细胞瘤U-87 MG、肾透明细胞腺癌细胞786-O、表皮癌细胞A-431和人原髓细胞白血病细胞HL-60的半数抑制浓度(IC_(50))范围为0.094~0.453 mg/mL,小于有效阈值0.5 mg/mL,即有效;其他4种佐太的浸提液尽管对某些肿瘤细胞增殖显示显著性抑制,但是未达到有效阈值。由此初步揭示了佐太5种前处理液对15种人源肿瘤细胞系抑制的差异,其中佐太DMSO悬液具有抗肿瘤潜能,为探索佐太体内抗癌活性提供了重要的体外实验数据支撑。The purpose of this research is to explore the in vitro inhibitory effect of classic Tibetan medicine Zuotai on human tumor cell lines.15 human tumor cell lines from 11 common clinical cancers(liver cancer,breast cancer,stomach cancer,colon cancer,ovarian cancer,cervical cancer,lung cancer,glioblastoma,kidney cancer,skin cancer,leukemia)were selected,and the inhibitory ratios of the five pretreatment solutions(DMSO suspension,the unfiltered extract of artificial gastric juice,the filtered extract of artificial gastric juice,the non-filtered extract of artificial intestinal juice,and the non-filtered extract of artificial juice extract)of Zuotai diluted in cell culture medium on tumor cell lines proliferation were determined,and the median inhibitory concentration(IC_(50)) was calculated.It was found that the Zuotai DMSO suspension diluted by cell culture medium shown significant effects on liver cancer cell lines(BEL-7402 and SMMC-7721),breast cancer cell lines(MDA-MB-231 and MCF-7),stomach cancer cell lines(MKN-45 and SGC-7901),colon cancer cell line HCT 116,ovarian cancer cell lines(HO-8910 and SK-OV-3),cervical cancer cell line Hela,lung cancer cell line A549,brain astroglioblastoma U-87 MG,primary renal clear cell adenocarcinoma 786-O,epidermal cancer cell A-431,and human promyelocytic leukemia cell HL-60 with a range of median inhibitory concentration from 0.094 to 0.453 mg/mL,which is less than the threshold value 0.5 mg/mL of the effective criteria for traditional medicine crude extracts against cancer.The other four extracts of Zuotai also shown inhibition on the proliferation of some tumor cells to some extent,but did not reach the effective threshold.In conclusion,this study has preliminarily revealed the difference in the inhibition of 15 human tumor cell lines by the five pretreatment solutions of Zuotai.Among them,Zuotai DMSO suspension has anti-tumor potential,which provides importantly in vitro experimental data for exploring the anti-cancer activity of Zuotai in vivo.
