蜈蚣败毒饮对银屑病模型鼠外周血中Treg细胞表达影响的相关性研究

摘　　要：目的以咪喹莫特诱导的银屑病鼠模型为研究对象,观察中药制剂蜈蚣败毒饮对其皮损指数及外周血中Treg细胞表达和其分泌因子白细胞介素10(interleukin-10,IL-10)、转化生长因子β(transforming growth factor-β,TGF-β)的干预作用,并对其之间相关性进行分析。方法将大鼠随机分为6组,分别为空白组、模型组、甲氨蝶呤西药对照组、蜈蚣败毒饮低、中、高剂量组,大鼠经咪喹莫特进行银屑病造模,再行不同组别的药物灌胃,8天后采用改良的鼠银屑病皮损面积和严重指数评分法(mice psoriasis area and severity index,MPASI)对银屑病鼠的皮损指数进行评价,并计算出灌胃前后MPASI的变化量(△MPASI),通过标记血细胞簇分化抗原4(cluster differentiation 4,CD4^(+))、血细胞簇分化抗原25(cluster differentiation 25,CD25^(+))、叉头框蛋白3(forkhead box p 3,Foxp3^(+))的T淋巴细胞(thymus,T),采用流式细胞法对外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中调节性T细胞(regulatory T cell,Treg)的表达量情况进行检测,采用酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)对外周血中IL-10和TGF-β进行检测,同时分别分析Treg细胞表达量、IL-10含量水平、TGF-β含量水平与模型灌胃前后△MPASI之间的相关性。结果(1)灌胃后各药物组银屑病鼠的MPASI分值均有所下降(P<0.05),尤以西药组和高剂量组的下降程度最为显著(P<0.05),且△MPASI的变化趋势与灌胃后MPASI相一致。(2)银屑病鼠外周血中Treg的细胞数均较正常鼠显著降低(P<0.05),西药组和高剂量组外周血中Treg的细胞数相对较高(P<0.05)。(3)西药组和高剂量组对于分泌因子IL-10、TGF-β的上调作用最为显著(P<0.05)。(4)经药物干预后,模型鼠外周血中Treg细胞的表达量、IL-10水平、TGF-β水平与△MPASI间均存在正相关性(P<0.05)。结论外周血中Treg细胞的表达量与银屑病鼠的皮损间存在着一定的关系,蜈蚣败�Objective The model of psoriasis in mice induced by imiquimod was used as the research object,to observe the intervention effect of Wugong Baidu Decoction on skin lesion index,the expression of Treg cell and its secretory factors interleukin-10(IL-10)and transforming growth factor-β(TGF-β)in peripheral blood,meanwhile,the correlation between them was analyzed.Methods The rats were randomly divided into 6 groups:the blank group,the model group,the methotrexate western medicine control group,the Wugong Baidu Decoction low,medium and high dose groups,the psoriasis model rats was established by imiquimod,and then different groups of drugs were administered by gavage,after 8 days,the modified mice psoriasis area and severity index(MPASI)scores were used to evaluate the skin lesion index of psoriatic mice,and the change of MPASI before and after gavage(△MPASI)was calculated,flow cytometry was used to detect the expression of regulatory T cell(Treg)in peripheral blood mononuclear cell(PBMC)by labeling thymus(T)with signals of cluster differentiation 4(CD4^(+)),cluster differentiation 25(CD25^(+)),forkhead box p 3(Foxp3^(+)),IL-10 and TGF-βin peripheral blood were detected by enzyme linked immunosorbent assay(ELISA),at the same time,the correlation between the expression of Treg cells,the content of IL-10 or TGF-βand△MPASI before and after gavage were analyzed respectively.Results(1)After intragastric administration,the skin lesion index of psoriasis rats in each drug group were decreased(P<0.05),in particular,western medicine group and the high dose group showed the most significant decrease(P<0.05),and the change trend of△MPASI was consistent with MPASI after gavage.(2)The number of Treg cells in peripheral blood of psoriatic rats was significantly lower than that in the normal mice(P<0.05),and the number of Treg cells in the peripheral blood of western medicine grou and the high dose group was relatively high(P<0.05).(3)Western medicine group and the high dose group had the most significant effects on up reg
